摘  要:为了丰富大叶黄精(Polygonatum kingianum var.grandifolium)根茎发育的转录组数据,发掘参与皂苷生物合成的功能基因,基于Illumina深度测序平台,对大叶黄精根茎进行转录组测序,并对次生代谢途径中皂苷的生物合成通路进行深入解析。结果显示,大叶黄精根茎的转录组数据经拼接后获得198296条转录本、92858条基因序列。基于超高效液相色谱和串联质谱(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)检测平台和自建数据库共检测到238种差异显著代谢物。根据大叶黄精皂苷合成途径中酶基因注释和差异基因的表达及代谢途径富集的分析,一共找到3条合成皂苷途径的相关通路以及33个酶基因,推测这些酶基因在皂苷合成途径中起关键作用。该研究为大叶黄精的转录组学和代谢组学研究提供了丰富的参考数据,对于大叶黄精的功能基因组学研究具有重要意义。In order to enrich the transcriptome data of the rhizome development of Polygonum kingianum var.grandifolium,and explore the functional genes involved in saponin biosynthesis,based on Illumina deep sequencing platform,this study conducted transcriptome sequencing on rhizomes of Polygonum kingianum var.grandifolium at seedling stage,and further analyzed the biosynthetic pathway of saponins in secondary metabolic pathway.The results showed that a total of 198296 transcripts and 92858 unigene sequences were obtained from the transcriptome data of rhizomes of Polygonum kingianum var.grandifolium by splicing,and 238 significantly different metabolites were detected based on the ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)detection platform and the self built database.According to the injection and differential gene expression of enzyme genes in the saponin synthesis pathway of Polygonum kingianum var.grandifolium and the enrichment analysis of metabolism pathway,three related pathways and enzyme genes in the saponin synthesis pathway were found,as well as the 33 enzyme-genes,and it was speculated that these enzyme genes played a crucial role in the saponin synthesis pathway.This study provides a wealth of reference data for the study of transcriptome and metabolomics of Polygonum kingianum var.grandifolium,and is of great significance for the study of functional genomics of Polygonum kingianum var.grandifolium.
