红山茶花、叶提取液在化妆品中的抗老功效研究

摘　　要：目的:通过体外测试探究红山茶花、叶提取液在化妆品中抗老(抗皱、抗氧化、抗糖化)的能力。方法:通过实时荧光定量PCR和ELISA试剂盒测试红山茶花、叶提取液在斑马鱼和成纤维细胞中的抗皱能力,使用流式细胞术和MTT比色法测试其在角质形成细胞的抗氧化能力,使用免疫组织化学测试其在离体皮肤中的糖基化终末产物含量评价抗糖化能力。结果:(1)红山茶花、叶提取液具有促进Ⅰ型胶原蛋白及抑制基质金属蛋白酶MMP-1和MMP-3的能力,可达到抗皱的效果;(2)可抑制活性氧和提升超氧化物歧化酶的含量,具有抗氧化的能力;(3)可抑制表皮层和真皮层中糖基化终末产物的表达,具有抗糖化的效果。结论:红山茶花、叶提取液可从抗皱、抗氧化和抗糖化方面达到抗老的效果。Objective:To explore the anti-aging(anti-wrinkle,anti-oxidation,anti-glycation)ability of Camewa japonica L.flower and leaf extract in cosmetics by in vitro testing.Methods:The anti-wrinkle ability of C.japonica flower and leaf extract in Brachydanio rerio and fibroblasts was tested by realtime PCR and ELISA kit,respectively.In addition,the antioxidant ability of C.japonica flower and leaf extract in keratinocytes was tested by flow cytometry and MTT colorimetry.What’s more,the anti-glycation of C.japonica flower and leaf extract in ex vivo skin was evaluated by immunohistochemistry.Results:(1)C.japonica flower and leaf extract had the ability to promote type I collagen and inhibit matrix metalloproteinases MMP-1 and MMP-3,achieving the ability of anti-wrinkle.(2)It inhibited reactive oxygen species and increase the content of superoxide dismutase,achieving the ability of anti-oxidation.(3)It inhibited the expression of advanced glycation end products in the epidermis and dermis,achieving the ability of anti-glycation.Conclusion:C.japonica flower and leaf extract had the ability of anti-aging from anti-wrinkle,anti-oxidation and anti-glycation.
