摘 要:目的:观察桃金娘小孢子不同发育时期的花器外部形态及细胞学特征,找出小孢子发育时期与花器相关性,为花药培养提供细胞学依据,同时探究低温预处理时间和植物生长调节剂组合对花药愈伤组织形成的影响。方法:用游标卡尺测量花蕾形态特征,用显微镜观察小孢子发育时期。接种前低温预处理花蕾0、12、24、48、72 h,采用L9(33)2,4-D(1.0、2.0、3.0 mg/L),6-BA(0.5、1.0、1.5 mg/L),NAA(0.1、0.2、0.3 mg/L)正交实验诱导花药愈伤组织。结果:小孢子处于单核靠边期时,花蕾横径为7.877±0.374 mm,纵径为7.167±0.340 mm,花瓣露出花萼,花药黄色,有浓烈芳香气味。4℃低温预处理花药24 h,诱导率为23.78%,显著高于其他处理。2,4-D是影响桃金娘花药愈伤组织形成的主要因子,配方MS+2.0 mg/L 2,4-D+1.5 mg/L 6-BA+0.1 mg/L NAA诱导率为30.82%,显著高于其他处理。结论:桃金娘小孢子各发育时期与花萼位置、花药颜色、花蕾大小密切相关,低温预处理可以提高花药愈伤组织诱导率,适宜的生长调节剂浓度和组合有利于提高花药愈伤组织诱导率。Objective:To observe the external morphological and cytological characteristics of Rhodomyrtus tomentosa microspores at different developmental periods of the floral apparatus,identify the correlation between microspore developmental periods and floral apparatus,provide cytological basis for anther culture,and explore the effects of low-temperature pretreatment time and plant growth regulator combinations on anther healing tissue formation.Methods:Measure the morphological characteristics of flow buds using vernier calipers and observe the developmental stage of microspores under a microscope.Low-temperature pretreatment of flower buds for 0,12,24,48 and 72 h before inoculation with L9(33)2,4-D(1.0,2.0,3.0 mg/L),6-BA(0.5,1.0,1.5 mg/L),NAA(0.1,0.2,0.3 mg/L)orthogonal experiments were conducted to induce anther healing tissues.Results:When the microspores were in the mononuclear leaning stage,the buds had a transverse diameter of 7.877±0.374 mm and a longitudinal diameter of 7.167±0.340 mm,with petals exposing the calyx and yellow anthers with a strong aromatic odor.24 h of low temperature pretreatment of anthers at 4°C resulted in a significantly higher induction rate of 23.78%than the other treatments.2,4-D was the main factor affecting the formation of myrtle anther healing tissues.The formula MS+2.0 mg/L 2,4-D+1.5 mg/L 6-BA+0.1 mg/L NAA induced 30.82%,which was significantly higher than the other treatments.Conclusion:The various developmental periods of R.tomentosa microspores were closely related to calyx position,anther color and bud size,low temperature pretreatment could improve the induction rate of anther healing tissue,and suitable growth regulator concentration and combination were beneficial to improve the induction rate of anther healing tissue.
