摘 要:目的 通过监测与分析河北省疾病预防控制中心脊髓灰质炎(简称“脊灰”)实验室2016-2021年使用的人横纹肌肉瘤(human rhabdomyosarcoma, RD)细胞系和转人脊灰病毒受体基因的鼠肺细胞(mouse L cells expressing the human Poliovirus receptor, L20B)系质量,确保检测脊灰病毒和非脊髓灰质炎肠道病毒(non-polio Enteroviruses, NPEV)的敏感性,为维持无脊灰状态提供可靠的技术保证。方法 采用聚合酶链反应(polymerase chain reaction, PCR)法对所用的RD细胞系和L20B系进行支原体检测(Mycoplasma detection);采用96孔微量培养板病毒滴定法对RD细胞系和L20B系进行细胞敏感性(cell sensitivity, CS)监测。结果 2016-2021年RD细胞系和L20B系支原体检测结果为阴性。与河北省脊灰实验室质量控制(laboratory quality control, LQC)株比较,RD细胞系和L20B系滴度波动范围为±0.5log CCID50/0.1ml, RD细胞系均值及标准差为Ⅰ型7.86±0.17,Ⅲ型7.73±0.13;L20B系均值及标准差为Ⅰ型7.05±0.10,Ⅲ型6.48±0.14。结论 2016-2021年河北省脊灰实验室使用的RD细胞系和L20B系质量控制良好,所用细胞系未受支原体污染,对Ⅰ型和Ⅲ型的脊灰病毒敏感性良好,为河北脊灰实验室脊灰病毒和非脊髓灰质炎肠道病毒分离及型别鉴定的质量提供了重要保障。Objective To ensure the sensitivity of the detection of Polioviruses and non-polio Enteroviruses(NPEV)and to provide reliable technical assurance for maintaining a polio-free status by monitoring and analyzing the quality of the human rhabdomyosarcoma(RD)cell line and mouse L cells expressing the human Poliovirus receptor(L20B)line used in the polio laboratory of Hebei Center for Disease Control and Prevention from 2016 to 2021.Methods Polymerase chain reaction(PCR)was used to detect Mycoplasma in the RD cell line and L20B line.The cell sensitivity(CS)of the RD cell line and L20B line was monitored by viral titration in 96-well microculture plates.Results From 2016 to 2021,the detection results of Mycoplasma in RD cell line and L2OB line were negative.Compared with the laboratory quality control(LQC)strain for polio in Hebei Province,the titers of the RD cell line and L20B line fluctuated in the range of+0.5log CCIDso/0.1ml.The mean and standard deviation of the RD cell line was 7.86+0.17 for type I and 7.73±0.13 for type II.The mean and standard deviation of the L20B line was 7.05+0.10 for type I and 6.48±0.14 for type II.Conclusion The quality control of the RD cell line and L20B line used in the polio laboratory in Hebei Province from 2016 to 2021 is good.The cell lines used are not contaminated by Mycoplasma and are good sensitivity to the type I and type II of Poliovirus,which provided an important guarantee for the quality of isolation and identification of Poliovirus and non-polio Enteroviruses in the polio laboratory in Hebei Province.
