摘 要:目的探讨白细胞介素-10(Interleukin-10,IL-10)基因启动子区单核苷酸多态性(single nucleotide polymorphisms,SNP)与儿童特应性皮炎(atopic dermatitis,AD)易感性之间的相关性。方法采用SNaPshot多重单碱基延伸反应检测IL-10基因启动子区rs1800896、rs1800871和rs1800872位点的SNP,分析各位点的基因型和等位基因频率与儿童AD易感性之间的相关性。结果共纳入儿童AD患者130例,同期健康体检儿童141例。rs1800896位点的基因型和等位基因频率在AD患者组和健康对照组之间差异有统计学意义(P=0.007,0.003);Logistic回归分析表明,具有AA基因型的个体患特应性皮炎的风险要显著高于GA基因型(P=0.003,OR=2.889,95%CI:1.447~5.769);具有A等位基因的个体患特应性皮炎的风险要显著高于G等位基因(P=0.003,OR=2.544,95%CI:1.364~4.745)。两组间rs1800871和rs1800872位点的基因型及等位基因频率差异均无统计学意义。三个SNP位点各基因型的临床特征比较差异均无统计学意义。结论IL-10基因启动子区rs1800896位点SNP可能与厦门及周边地区儿童AD的易感性相关。Objective To assess the association between polymorphisms of interleukin⁃10(IL⁃10)gene promoter and the susceptibility to atopic dermatitis(AD).Methods The single nucleotide polymorphisms(SNPs)of IL⁃10 gene promoter were detected by single base extension(SNaPshot)assay.Results One hundred and thirty AD patients and 141 normal control were enrolled.There were statistical differences between AD patients and normal control in genotype and allele distribution at rs1800896(P=0.007,0.003).Subjects with AA genotype had distinctly increased risks for AD than subjects with GA genotype(P=0.003,OR=2.889,95%CI:1.447⁃5.769),and subjects with A allele had distinctly increased risks for AD than subjects with G allele(P=0.003,OR=2.544,95%CI:1.364⁃4.745).There were no significant differences in genotype and allele frequencies of rs1800871 and rs1800872 between AD patients and normal controls.There were no significant differences in the clinical characteristics of each genotype of the three SNPs.Conclusion This study shows that SNP atrs1800896 of IL⁃10 gene promoter may be associated with susceptibility to AD in Xiamen and peripheral area.
