摘  要:【目的】BLTY品系辣椒叶片对1~3龄斜纹夜蛾幼虫有较强的胃毒性,本研究旨在进一步探明这种胃毒性与肠道细菌群落结构及多样性的相关性。【方法】利用Illumination HiSeq技术对斜纹夜蛾3龄幼虫取食BLTY品系辣椒叶片、FXBX辣椒叶片(无毒性)及人工饲料后肠道细菌16S rRNA的特异性区域进行测序,应用Usearch和R语言软件分析其肠道细菌群落结构、物种丰度及多样性的差异。【结果】厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)为不同处理下斜纹夜蛾3龄幼虫肠道细菌的共有优势菌门。相较于取食无毒性的FXBX辣椒叶片和人工饲料,取食BLTY品系辣椒叶片的肠道菌群Alpha多样性指数显著上升,菌群组成和结构差异显著。在门水平上,变形菌门、放线菌门(Actinobacteria)和拟杆菌门(Bacteroidetes)相对丰度升高;在属水平上,有益菌肠球菌属(Enterococcus)相对丰度降低,而致病菌沙门氏菌属(Salmonella)、布鲁氏菌属(Brucella)、寡养单胞菌属(Stenotrophomonas)相对丰度显著升高。KEGG代谢差异分析表明,取食BLTY品系辣椒叶片会使肠道细菌外源化合物的生物降解和代谢通路的相对丰度显著上升。【结论】斜纹夜蛾在取食BLTY品系辣椒叶片后肠道细菌群落结构和多样性发生变化,即有益菌的相对丰度下降,而致病菌的相对丰度上升。【Objective】The study aimed to study the relationship between the diversity of gut microbiota in Spodoptera litura larvae and leaves of pepper line BLTY,a line known to exert gastric toxicity to S.litura 1st-3rd larvae.【Method】S.litura 3rd larvae were analyzed on community structure,species richness and diversity of gut microbiota by 16S rDNA sequencing(Illumina HiSeq high-throughput sequencing technology)followed by Usearch and R Programming after fed with leaf of pepper BLTY,leaf of FXBX(non-toxicity pepper line)or artificial diet.【Result】Firmicutes and Proteobacteria were the common dominant phylum in the guts of S.litura 3rd larvae fed with different diets.Compared with larvae fed with leaves of FXBX or artificial diet,the gut microbiota of larvae fed with leaf of BLTY had significantly increased Alpha diversity index and different bacterial composition and structure.They witnessed increased relative abundances of Proteobacteria,Actinobacteria and Bacteroidetes at the phylum level,significantly increased relative abundances of pathogenic Salmonella,Brucella and Stenotrophomonas but decreased relative abundance of beneficial bacteria Enterococcus at the genus level.KEGG metabolic analysis revealed that intake of leaf of BLTY was contributed to significantly higher relative abundance of xenobiotics biodegradation and metabolism pathway in the gut microbiota of larvae.【Conclusion】The composition and diversity of gut microbiota in S.litura changed after fed with leaf of BLTY,that is,declined relative abundance of beneficial bacteria while increased relative abundance of pathogenic bacteria.
