摘  要:目的:观察健脾养正方联合紫杉醇节律化疗对小鼠胃癌肺转移的抑制作用,及对小鼠紫杉醇化疗所致的骨髓抑制和免疫损伤的影响。方法:30只615品系小鼠随机分为模型组、常规剂量的紫杉醇化疗组(PTX)、紫杉醇节律化疗组(MC-PTX),健脾养正方组(JPYZ),健脾养正方联合紫杉醇节律化疗组(MC-PTX+JPYZ),每组6只,小鼠尾静脉注射MFC细胞建立胃癌肺转移模型。给药2周后观察肿瘤浸润肺组织情况,计算肺转移抑制率;完整剥离脾、胸腺组织,计算胸腺指数、脾指数;HE染色观察骨髓病理组织学改变及骨髓细胞占骨髓腔比值;流式细胞仪检测骨髓细胞凋亡情况;血细胞分析检测外周血中白细胞、红细胞、血小板、血红蛋白的改变。结果:1.MC-PTX+JPYZ组肿瘤浸润及肺转移灶结节最少且抑瘤率最高(73.14%),显著高于其它组(P<0.05);2.与PTX组和MC-PTX组比较,MC-PTX+JPYZ组骨髓细胞占骨髓腔比值显著升高(P<0.05),骨髓细胞的早期凋亡率和总凋亡率降低明显,升高了紫杉醇化疗后骨髓抑制小鼠的白细胞水平(P<0.05)且对骨髓有明显的促恢复作用。3.与PTX组和MC-PTX组相比,MC-PTX+JPYZ组体质量、胸腺、脾指数均上升(P<0.05)。结论:健脾养正方联合紫杉醇节律化疗能显著增加紫杉醇治疗胃癌肺转移的疗效,显著减轻紫杉醇引起的骨髓抑制及免疫功能损伤。Objective:To observe the inhibitory effect of Jianpi Yangzheng formula combined with paclitaxel metronomic chemotherapy on lung metastasis of gastric cancer in mice,and its effect on bone marrow suppression and immune injury induced by paclitaxel chemotherapy.Methods:Thirty 615 strain mice were randomly divided into model group,conventional dose of paclitaxel chemotherapy group(PTX),paclitaxel metronomic chemotherapy group(MC-PTX),and Jianpi Yangzheng formula group(JPYZ),Jianpi Yangzheng formula combined with paclitaxel metronomic chemotherapy group(MC-PTX+JPYZ),with 6 mice in each group.MFC cells were injected into the tail vein of the mice to establish lung metastasis model of gastric cancer.After 2 weeks of administration,tumor infiltration of lung tissue was observed,and inhibition rate of lung metastasis was calculated.The spleen and thymus tissues were completely stripped,and the thymus index and spleen index were calculated.HE staining was used to observe the histopathological changes of bone marrow and the ratio of bone marrow cells to bone marrow cavity.The apoptosis of bone marrow cells was detected by flow cytometry.The changes of white blood cells,red blood cells,platelets and hemoglobin in peripheral blood were detected by blood cell analysis.Results:1.MC-PTX+JPYZ group had the least tumor infiltration and pulmonary metastasis nodules,and the highest tumor inhibition rate(73.14%),which was significantly higher than other groups(P<0.05).2.Compared with PTX and MC-PTX groups,the ratio of bone marrow cells to bone marrow cavity in MC-PTX+JPYZ group was significantly increased(P<0.05),the early apoptosis rate and total apoptosis rate of bone marrow cells were significantly decreased,the white blood cell level of myelosuppression mice after paclitaxel chemotherapy was increased(P<0.05),and the bone marrow function was significantly promoted.3.Compared with PTX and MC-PTX groups,the body weight,thymus and spleen index of MC-PTX+JPYZ group were increased(P<0.05).Conclusion:Jianpi Yangzheng formula combined
