摘  要:目的:观察艾灸“足三里”对亚急性衰老模型大鼠氧化应激和肠道菌群的影响,探讨艾灸延缓衰老的可能作用机制。方法:将30只SD大鼠随机分为空白组、模型组、足三里组,每组10只。模型组、足三里组大鼠按500 mg/kg剂量腹腔注射D-半乳糖制备亚急性衰老模型,每日1次,连续42 d。足三里组艾灸双侧“足三里”,每日1次,每次每穴3壮,连续28 d。采用ELISA法检测各组大鼠干预后血清超氧化物歧化酶(SOD)及丙二醛(MDA)含量;采用16S rRNA测序检测各组大鼠肠道菌群。结果:与空白组比较,模型组大鼠血清SOD含量减少(P<0.01),MDA含量增加(P<0.01);与模型组比较,足三里组大鼠血清SOD含量增加(P<0.01),MDA含量减少(P<0.01)。与空白组比较,模型组大鼠Chao1、Shannon指数降低(P<0.01,P<0.05);与模型组比较,足三里组大鼠Chao1、Shannon指数升高(P<0.01,P<0.05)。与空白组比较,模型组大鼠厚壁菌门、密螺旋体属_2及毛螺菌科NK4A136相对丰度升高(P<0.05,P<0.01),普雷沃氏菌科UCG-003相对丰度及拟杆菌门与厚壁菌门相对丰度比值(B/F值)降低(P<0.01,P<0.05);与模型组比较,足三里组厚壁菌门、密螺旋体属_2相对丰度降低(P<0.01),拟杆菌门、乳杆菌属、普雷沃氏菌科UCG-003相对丰度及B/F值升高(P<0.05,P<0.01)。结论:艾灸“足三里”可有效改善衰老大鼠氧化应激水平,调节肠道菌群结构,维持肠道菌群微生态平衡,从而起到延缓衰老的作用。Objective To observe the effects of moxibustion at"Zusanli"(ST 36)on oxidative stress and intestinal flora in subacute aging rats,and to explore the possible mechanism of moxibustion in delaying aging.Methods Thirty SD rats were randomly divided into a blank group,a model group and a Zusanli group,with 10 rats in each group.Subacute aging model was established by intraperitoneal injection of D-galactose at dosage of 500 mg/kg in the model group and the Zusanli group,once a day for 42 days.In the Zusanli group,moxibustion was applied at bilateral"Zusanli"(ST 36),once a day,3 moxa cones at each acupoint,for consecutive 28 days.After intervention,the serum levels of superoxide dismutase(SOD)and malondialdehyde(MDA)were detected by ELISA;the intestinal flora was detected by 16S rRNA sequencing technique in each group.Results Compared with the blank group,the serum level of SOD was decreased(P<0.01),the serum level of MDA was increased(P<0.01)in the model group.Compared with the model group,the serum level of SOD was increased(P<0.01),the serum level of MDA was decreased(P<0.01)in the Zusanli group.Compared with the blank group,Chao1 and Shannon indexes were decreased in the model group(P<0.01,P<0.05).Compared with the model group,Chao1 and Shannon indexes were increased in the Zusanli group(P<0.01,P<0.05).Compared with the blank group,the relative abundance of Firmicutes,Treponema_2 and Lachnospiraceae_NK4A136_group was increased(P<0.05,P<0.01),while the relative abundance of Prevotellaceae_UCG-003 and the relative abundance ratio of Bacteroidetes and Firmicutes(B/F value)were decreased(P<0.01,P<0.05)in the model group.Compared with the model group,the relative abundance of Firmicutes and Treponema_2 was decreased(P<0.01),while the relative abundance of Bacteroidetes,Lactobacillus,Prevotellaceae_UCG-003 and B/F value were increased(P<0.05,P<0.01)in the Zusanli group.Conclusion Moxibustion at"Zusanli"(ST 36)can effectively improve the level of oxidative stress,regulate the constitution of intestinal flora,maintain the
