摘  要:圆齿野鸦椿是具有极高观赏和药用开发价值的常绿小乔木。在热胁迫响应中,热激转录因子(heat shock transcription factor, Hsf)可以与热激蛋白(heat shock protein, Hsp)基因上游启动子相结合而被鉴定,是植物生长发育和响应环境胁迫的重要调控因子,对植物的生长发育十分重要。Hsf基因家族在许多植物中已被鉴定,但在圆齿野鸦椿中的功能尚不清楚,本研究对圆齿野鸦椿Hsf基因家族进行了鉴定与生物信息学分析。结果表明,圆齿野鸦椿基因组编码有22个Hsf家族基因,根据它们的蛋白质结构和系统发育特征可以分为3组。基因结构和结构域分析表明Hsf基因在进化过程中相对保守。顺式作用元件分析表明EkHsf基因在圆齿野鸦椿应答各种胁迫过程中发挥重要作用。共线性分析结果表明,片段复制事件对圆齿野鸦椿Hsf基因家族的进化和扩张起着重要作用。EkHsf基因在不同组织和发育阶段的表达显示出组织特异性。最后通过共表达分析鉴定出EkHsf-B4b和EkHsf-B1b为潜在圆齿野鸦椿花青素合成的调控因子。本研究结果为进一步研究圆齿野鸦椿Hsf基因的生物学功能提供科学依据。The Euscaphis konishii is small evergreen trees with high medicinal and ornamental value.Heat shock transcription factor(Hsf)acts as activators of heat shock protein(Hsp)and plays multiple roles in response to environmental stress and plant development.Although the Hsf gene family has been identified in many plants,it is unknown in E.konishi.This study was conducted to identify and bioinformatically analyze the E.konishi Hsf gene family.The results showed that there were 22 Hsf family genes in E.konishi,which could be classified into three groups based on their protein structures and phylogenetic characteristics.Gene structure and structural domain analysis indicated that Hsf genes were relatively conserved during evolution.Analysis of cis-acting elements showed that EkHsf genes play important roles in the response to various stresses in E.konishi.The results of synteny analysis indicated that fragment replication events played an important role in the evolution and expansion of the Hsf gene family in E.konishi.The expression of EkHsf genes in different tissues and developmental stages showed tissue specificity.Finally,EkHsf-B4b and EkHsf-Blb were identified by co-expression analysis as potential regulators of anthocyanin synthesis in E.konishi.The results of this study provide a scientific basis for further study on the biological function of Hsf gene of E.konishi.
