摘  要:目的建立测定家种连翘中山柰酚含量的高效液相色谱(HPLC)法。方法色谱柱为Shim-pack VP-ODS柱(250 mm×4.6 mm,4.6μm),流动相为甲醇-0.4%磷酸水溶液(50∶50,V/V),流速为1.0 mL/min,检测波长为365 nm,柱温为30℃,进样量为10μL。结果山柰酚质量浓度在0.01~0.5 mg/mL范围内与峰面积线性关系良好;精密度、稳定性、重复性试验结果的RSD均小于5%;平均加样回收率为97.30%,RSD为2.13%(n=6);湖北省十堰市郧西县8个村家种连翘中山柰酚含量为0.0377~1.0887 mg/g。结论该方法灵敏、可重复,测定结果准确,可用于家种连翘中山柰酚含量的测定。Objective To establish a high-performance liquid chromatography(HPLC)method for content determination of kaempferol in domesticated Forsythiae Fructus.Methods The chromatographic column was Shim-pack VP-ODS column(250 mm×4.6 mm,4.6μm),the mobile phase was methanol-0.4%phosphoric acid aqueous solution(50∶50,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 365 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear range of kaempferol was 0.01-0.5 mg/mL.The RSDs of precision,stability and repeatability tests were all lower than 5%.The average recovery rates of kaempferol was 97.30%,with the RSD of 2.13%(n=6).The content of kaempferol in domesticated Forsythiae Fructus from eight villages of Yunxi County,Shiyan City,Hubei Province was in the range of 0.0377 to 1.0887 mg/g.Conclusion This method is sensitive,repeatable and accurate,which can be used for the content determination of kaempferol in domesticated Forsythiae Fructus.
