不同处理方式下木霉真菌降解昭通褐煤的差异性分析

摘  要:真菌对褐煤的降解作用明显,为研究不同处理方式下真菌对褐煤降解作用的差异,利用污水处理厂活性污泥中分离出的一株木霉真菌H-2,通过工业分析、紫外/可见光全波长扫描、FT-IR和HPLC-QTOF/MS等技术手段,研究了该菌株在菌/煤固态培养、菌/煤液态同时培养、菌/煤液态差时培养三种不同处理方式下对云南昭通褐煤的降解特性。结果表明:3种处理方式下,真菌H-2对煤中化学键断裂和大分子结构解聚的作用明显,作用后煤中羟基、甲基、亚甲基等官能团增多;固态培养条件下,褐煤经真菌H-2作用后,表面出现明显裂隙,结构疏松,易松散破碎,而菌/煤液态差时培养处理方式下,降解产物更易被解离至培养液中,主要为芳香化合物、不饱和的杂环化合物和少量不饱和的醛酮类化合物。可见,真菌H-2的生物降解作用对褐煤的物化性质产生了影响,并且不同处理方式下煤的生物降解产物也存在差异。Fungi have obvious degradation effect on lignite.In order to study the difference of fungi degradation effect on lignite under different treatment methods,a Trichoderma strain H-2 isolated from activated sludge of sewage treatment plant was used to study its degradation characteristics of Yunnan Zhaotong lignite under three different conditions of bacteria and coal solid culture,bacteria and coal liquid simultaneous culture and bacteria and coal liquid differential culture by means of industrial analysis,UV/visible full wavelength scanning,FT-IR and HPLC-QTOF/MS.And the following research results are obtained:under three different conditions,strain H-2 has an obvious effect on the breaking of chemical bonds and the depolymerization of macromolecular structure in lignite,and the functional groups such as hydroxyl,methyl and methylene in coal increase after the action;under the treatment of solid culture,obvious cracks appear on the surface,the structure is loose,and it is easy to loosen and break after the action of strain H-2 on lignite,but under the treatment of liquid differential culture,lignite is more easily affected by strain H-2,and more degraded substances are dissociated into the culture medium,mainly aromatic compounds,unsaturated heterocyclic compounds and a small amount of unsaturated aldehydes and ketones.It can be seen that the biodegradation of strain H-2 has an impact on the physicochemical properties of lignite,and the biodegradation products of coal under different treatment methods are also different.
