拉氏鱥脊椎骨中TGF-β超家族部分基因的温度表达模式研究

摘  要:为探究拉氏鱥(Phoxinus lagowskii)在不同饲养温度下脊椎骨中与骨发育和生长有关基因的表达差异,设置4个温度组(15、20、25、30℃),以15℃为对照,饲养28 d后,统计分析饲养前后各组体长和体质量的变化,并提取脊椎骨的总RNA。随后基于比较转录组筛选出的差异表达基因,利用实时荧光定量PCR技术检测bmp4、bmp2b、bmp7a、smad4、smad7、gdf8和sox9a基因的相对表达量。结果显示,各组内饲养前后的平均体长和体重均无显著性变化;各组饲养前后的平均体长无组间差异,平均体质量在20℃和15℃组间没有显著差异,但25℃和30℃组的显著升高,且体质量的增长随着温度的升高而增加。bmp4的相对表达量呈现出明显的先上升后降低的趋势;bmp2b在25℃和30℃的表达量与对照组相比显著降低;bmp7a的表达量随温度的升高有上升趋势,25℃和30℃组显著高于对照组和20℃组。smad4、smad7和sox9a的变化趋势相似,从15℃升到25℃时表达量增多,水温升高到30℃时,表达量下降。gdf8在15℃时被显著抑制,温度升高,表达量明显增多。综合本研究结果可初步推测,拉氏鱥在20~25℃时生长状况较为良好,脊椎骨中骨形态发育和生长相关基因的表达处于相对较优的模式,可能为其更好的生长发育提供基础。The fish,an ectothermic organism inhabiting aquatic environments,is widely acknowledged for its thermosensitivity.Temperature is a crucial environmental factor that significantly influences fish growth,development,reproduction,and various other physiological activities.The elevation of temperature can enhance the metabolic activities of organisms.However,it will hamper their growth beyond a certain threshold and even induce organismal mortality.Phoxinus lagowskii has a wide distribution range in China,from the Yangtze River in the south to the Heilongjiang River in the north.However,due to geographical differences,there are variations in environmental temperatures among populations,resulting in divergent growth and development patterns.Therefore,it is crucial to investigate the relative molecular mechanisms of P.lagowskii under different temperature conditions.In this study,we explored the impact of temperature on the expression of genes associated with bone morphogenesis and growth of P.lagowskii.The individuals were reared in four separate groups at temperatures of 15,20,25,and 30℃for four weeks.The results indicated no statistically significant differences in standard length among all groups.Additionally,there was no significant difference in body weight between the 15℃and 20℃groups.However,the body weights of the 25℃and 30℃groups were significantly higher compared to the former two groups.The total RNA from vertebrae was extracted and reverse transcribed for qPCR analysis to determine the relative expression levels of bmp4,bmp2b,bmp7a,smad4,smad7,gdf8 and sox9a genes.The relative expression of bmp4 exhibited a significant increase from 20℃to 25℃,followed by a considerable decrease from 30℃,while remaining significantly higher than that of the control group(15℃).The expressions of bmp2b at 25℃and 30℃were significantly lower than that of the control group.Whereas,the expressions of bmp7a were the highest at 25℃and 30℃.The expression levels of smad4,smad7 and sox9a exhibited upward tren
