五加生化胶囊对药物流产出血模型大鼠Treg/Th17细胞平衡的影响及机制研究

摘　　要：目的 探究五加生化胶囊调控JAK2/STAT3通路药物流产出血模型大鼠Treg/Th17细胞平衡的影响。方法 选用雌性成年未孕SD大鼠24只,雄性成年大鼠12只,按雌雄比2∶1合笼。将受孕大鼠随机分为对照组(Control)、模型组(Model)和五加生化组(WSC),每组6只,建立异常子宫出血模型,测量各组大鼠子宫出血量;ELISA检测子宫组织匀浆中lL-10、IL-17A、TGF-β1、雌激素和孕激素水平;流式细胞术检测大鼠子宫组织中Treg、Th17细胞比例变化;Western blot检测大鼠子宫组织中JAK2、STAT3及其磷酸化蛋白的表达。结果 正常受孕组大鼠无阴道出血情况;Model组大鼠阴道有出血,血清中雌激素和孕激素水平均下降;大鼠血清中IL-17水平均增加,而IL-10和TGF-β1水平下降,子宫组织中Treg/Th17细胞比值下降,p-JAK2和p-STAT3蛋白表达增多。与模型组相比,五加生化组大鼠阴道出血量减少,血清中雌激素和孕激素水平增加,IL-17水平均减少,IL-10和TGF-β1水平增加,子宫组织中Treg/Th17细胞比值增加,p-JAK2和p-STAT3蛋白下降。结论 五加生化通过调控JAK2/STAT3信号通路维持药物流产大鼠Treg/Th17细胞的平衡。Objective To explore the effect of WujiaShenghua capsule on regulating the balance of Treg/Th17 in rats with drug abortion hemorrhage by JAK2/STAT3 pathway.Methods Twenty-four female adult non-pregnant SD rats and 12 male adult rats were selected,and cage the rats in the ratio of 2∶1.Pregnant rats were randomly divided into control group(Control),model group(Model),and WujiaShenghua group(WSC),with 6 rats in each group,established abnormal uterine bleeding model and measured the amount of uterine bleeding in each group of rats.The levels of lL-10,IL-17A,TGF-β1,estrogen and progestogen in uterine tissue homogenate were detected by ELISA.The changes in the proportion of Treg/Th17 cells in the rat uterine tissue were detected using flow cytometry.Western blot was used to detect the expression of JAK2,STAT3,and their phosphorylated proteins.Results There was no vaginal bleeding in the normal pregnant group,but there was bleeding in the model group,and the levels of estrogen and progestogen in serum were decreased.The level of IL-17 in serum of rats was increased,while the levels of IL-10 and TGF-β1 were decreased,the Treg/Th17 cell ratio was decreased in uterine tissue,and the expression of p-JAK2 and p-STAT3 were increased.Compared with the model group,the rats of WSC showed a decrease in vaginal bleeding,an increase in serum levels of estrogen and progestogen,a decrease in IL-17 levels,and a decrease in IL-10 and TGF levels-β,the Treg/Th17 cell ratio in uterine tissue was increased,and the p-JAK2 and p-STAT3 proteins were decreased.Conclusion WujiaShenghua capsule maintained the balance of Treg/Th17 cells in drug induced abortion rats by regulating the JAK2/STAT3 signaling pathway.
