摘　　要：目的 明确九香虫水煎液对小鼠的急性毒性,初步评价其安全性,为安全用药提供依据。方法 根据《中药、天然药物急性毒性研究的技术指导原则》([Z]GPT2-1),采用最大耐受量法观察九香虫水煎液对小鼠的急性毒性。将昆明小鼠随机分为对照组和给药组,每组20只,雌雄各半;连续灌胃14 d,每天1次;对照组给予生理盐水,给药组给予20 g/kg九香虫水煎液;观察小鼠中毒症状和活动反应,每2天称量小鼠体重,进食进水量。14 d后解剖小鼠,称量器官重量并进行HE染色,收集血清检测丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、肌酐(Cre)、尿素氮(BUN)指标。结果 与对照组相比,实验组小鼠无中毒症状和异常活动反应,体重、进食量和进水量、各器官脏器系数、血液ALT、AST、Cre、BUN指标无显著差异。给药后小鼠肝、脾、肾、胃、睾丸、卵巢未见病理性异常。结论 九香虫水煎液MTD(最大耐受剂量)>20 g/kg,无致死作用,具有较高的安全性。Objective To determine the acute toxicity of Aspongopus chinensis decoction(ACD)on mice,and to evaluate its safety,so as to provide evidence for safe drug use.Methods According to the Technical Guiding Principle of Acute Toxicity Study of Traditional Chinese Medicine and Natural Medicine([Z]GPT2-1),the acute toxicity of the decoction was observed by maximum tolerance dose method.Kunming mice were randomly divided into two groups:control group and drug administration group,with 20 mice in each group,half male and half female.For 14 days,the control group was given normal saline(NS)and the administration group was given ACD(20 g/kg).The poisoning symptoms and reaction of mice was observed.The weight of mice and the feed and water consumption was recorded every two days.Then the mice were dissected,the organs were weighed and HE staining was performed,and serum was collected for the detection of ALT,AST,Cre,BUN indexes.Results Compared with the control group,the experimental group had no toxic symptoms and abnormal reaction,and there were no significant differences in body weight,food and water intake,organ coefficients,ALT,AST,Cre,and BUN.No pathological abnormalities were observed in liver,spleen,kidney,stomach,testis,and ovary after administration.Conclusion The MTD(maximum tolerance dose)of ACD was>20 g/kg,which had no lethal effect with high safety.
