摘 要:为了挖掘番茄(Solanum lycopersicum)株高发育调控相关基因,本文以樱桃番茄Micro Tom及其矮化突变体dwt为材料,通过转录组测序和表达水平分析筛选鉴定到15个与株高发育调控相关的候选基因。其中,TCH4-like1、TCH4-like2、ABAR7、ABAR10、PP2C7、PP2C13和PR1等基因的表达异常,从而影响BRs、ABA和SA信号途径,并抑制番茄的生长,进而导致dwt番茄矮化。同时,ABAR7、ABAR10、PP2C7、PP2C13、PR1等基因与WRKY33、MPK3基因共同影响MAPK信号途径,从而调控dwt突变体矮化性状的形成。另外,peroxidase-like1、peroxidase-like2和peroxidase-like3等基因的表达受到强烈抑制,从而影响dwt木质素合成,并促成dwt的矮化特征形成。结果可为番茄株高发育调控中关键调控基因的鉴定奠定良好基础,同时也为番茄矮化育种提供重要线索。Taking Mrico Tom and dwt as materials in this study,l5 genes associated with the regulation of plant height development are screened out by transcriptome sequencing and expression level analysis in order to explore the genes related to the regulation of plant height development of tomatoes(Solanum lycopersicum).Among them,the abnormal expression of TCH4-likel,TCH4-like2,ABAR7,ABAR1O,PP2C7,PP2C13,and PRI could affect the BRs,ABA,and SA signaling pathway and inhibit the growth of tomatoes,leading to dwarfism in dwt.Meanwhile,ABAR7,ABAR10,PP2C7,PP2C13 and PRI,together with WRKY33 and MPK3,would affect the MAPK signaling pathway,regulating the formation of dwarfism mutant dwt.In addition,the expression level of peroxidase-likel,peroxidase-like2 and peroxidase-like3 is strongly inhibited,thus affecting lignin synthesis and promoting the dwarfism in dwt.These results will lay a good foundation for the identification of key regulatory genes in regulating the plant height development of tomatoes,and provide important clues for tomato dwarf breeding as well.
