摘 要:为探索延胡索治疗血栓的潜在作用机制,检索中药系统药理学数据库(TCMSP),对延胡索药效成分进行筛选,利用PubChem数据库获得药效成分3D结构,并通过Pharmmapper数据库筛选出各活性成分可能存在的靶点;通过CTD数据库挖掘与血栓相关的作用靶点,利用STRING获得与血栓相关的作用靶点的蛋白相互作用网络,通过GO功能富集和KEGG通路分析延胡索干预血栓的分子机制。结果表明:从延胡索中共筛选出13个主要成分,预测出延胡索与血栓相关靶点238个,选出26个关键靶点。GO富集分析结果主要涉及各种功能蛋白的活性、调控等过程。KEGG通路富集得到4条通路,包括蛋白聚糖在癌症中的应用、前列腺癌、膀胱癌以及丁酸代谢通路。综上所述,延胡索发挥抗血栓作用可能通过多成分、多靶点起作用,为延胡索药材质量标准的制订和提升提供了参考。The pharmacodynamic components of Corydalis yanhusuo are screened by searching the TCM Systematic Pharmacology Database(TCMSP)in order to explore the potential action mechanism of C.yanhusuo in the treatment of thrombus.PubChem database is employed to obtain the 3D structure of pharmacodynamic components and Pharmmapper database is utilized to screen out the potential targets of each active component.The target associated with thrombus is mined through CTD database and the protein interaction network(PPI)of the target associated with thrombus is obtained by STRING.The molecular mechanism of C.yanhusuo in intervention of thrombus is analyzed by GO functional enrichment and KECG pathway.The results show that a total of 13 main components are screened from C.yanhusuo and 238 targets related to thrombus are predicted,including 26 key targets screened.The results of GO enrichment analysis have mainly involved the activity and regulation of various functional proteins.KEGG pathway is enriched to obtain 4 pathways,including the application of proteoglycans in cancer,prostate cancer,bladder cancer and butanoate metabolic pathways.In conclusion,the antithrombotic effect of C.yanhusuo may be achieved through multi-components and multi-targets,providing references for the formulation and improvement of the quality standard of C.yanhusuo.
