摘 要:目的:探讨甲状腺功能减退与肾功能改变的因果关联。方法:使用两样本孟德尔随机化(MR)分析方法,将甲状腺功能减退显著相关的单核苷酸多态性(SNPs)作为工具变量,进行甲状腺功能减退与肾功能因果关联的探讨。数据来源全基因组关联研究的汇总数据。结果:共29个SNPs纳入为工具变量。甲状腺功能减退与肾小球滤过率之间无明显因果关联:逆方差加权(OR=2.33,95%CI:0.03,7.88)、MR-Egger回归(OR=0.11,95%CI:0.03,3.11)、加权中位数(OR=0.62,95%CI:0.06,7.91)、简单模式(OR=1.55,95%CI:0.12,15.3)及加权模式(OR=0.77,95%CI:0.11,9.03)(均P>0.05)。留一法敏感性分析和Cochrane’s Q检验提示所得结果稳健且不存在异质性(P>0.05)。结论:甲状腺功能减退与肾功能改变不存在因果关联。Objective:To investigate the causal relationship between hypothyroidism and renal function.Methods:Two-sample Mendelian randomization(MR)analysis was used to investigate the causal association between hypothyroidism and renal function,with single nucleotide polymorphisms(SNPs)significantly associated with hypothyroidism as instrumental variables.Data were collected from genome-wide association studies.Results:A total of 29 SNPs were included as instrumental variables.There is no significant causal correlation between hypothyroidism and estimated glomerular filtration rate:inverse variance weighted(OR=2.33,95%CI:0.03,7.88),MR-Egger regression(OR=0.11,95%CI:0.03,3.11),weighted median estimator(OR=0.62,95%CI:0.06,7.91),simple model(OR=1.55,95%CI:0.12,15.3),and the weighted model(OR=0.77,95%CI:0.11,9.03).Additionally,leave-one-out sensitivity analysis and Cochrane's Q test suggested robust results with no heterogeneity(P>0.05).Conclusion:There is no causal correlation between hypothyroidism and renal function.
