摘 要:目的制备羧甲司坦微囊,优选羧甲司坦微囊的制备工艺。方法采用复凝聚法,以明胶、阿拉伯胶为囊材制备羧甲司坦微囊。以包封率和载药率作为制剂的主要评价指标,通过单因素考察实验优选羧甲司坦微囊的制备工艺。结果羧甲司坦、明胶和阿拉伯胶组成的混合囊材的最适宜比例为1∶1∶1.5;最适宜的成囊温度为35℃;最适宜的搅拌速度为600 r/min;最适宜的pH为3.0;最适宜的固化剂添加量为2 mL。根据最优制备工艺制备三批羧甲司坦微囊,包封率分别为80.73%、83.39%、82.23%;载药量分别为42.15%、43.46%、41.41%。所制备的微囊在显微镜下观察,其形状规则呈类圆形,粒径约为5~25μm。结论本研究所确定的羧甲司坦微囊制备方法简便易操作,微囊粒径较小、圆整度好。Objective To prepare carbocisteine micro-capsules,and optimize the preparation process of carbocisteine sustained-release micro-capsules.Methods Carbocisteine micro-capsules were prepared using gelatin and gum acacia as the capsule material by composite coacervation,using encapsulation efficiency and loading efficiency as the main evaluation indexes,the preparation process of carbocisteine micro-capsules was optimized by single factor experiment.Results Carbocisteine micro-capsules were prepared using gelatin and gum acacia as the capsule material by composite coacervation.The optimum ratio of carbocisteine to gelatin and gum acacia was 1∶1∶1.5.The optimum forming micro-capsules temperature was 35℃.The optimum stirring speed was 600 r/min,the optimum pH was 3.0 and the optimum volume of curing agent was 2.0 mL.According to the optimal preparation process,the encapsulation efficiency of three batches of carbocisteine micro-capsules were 80.73%,83.39%and 82.23%,respectively.And loading efficiency were 42.15%,43.46%and 41.41%,respectively.The micro-capsules prepared by the selected preparation process were observed under a microscope with regular shape and particle size ranging from 5μm to 25μm.Conclusion The preparation process of carbocisteine micro-capsules was simple,micro-capsules were small particle size and good roundness.
