摘  要:试验旨在比较研究凝结芽孢杆菌培养液(B.coagulans culture solution,BCS)及其离心上清液(centrifuged supernatant of B.coagulans culture solution,CSS)、菌悬液(centrifuged bacterial suspension of B.coagulans culture solution,CBS)在不同pH条件下的抑菌性能差异。试验设计4个处理组:无菌蒸馏水组(sterile distilled water,SDW组,对照组)、BCS组、CSS组、CBS组。采用牛津杯体外抑菌试验法,参照畜禽胃肠道pH,检测4组在不同pH(2.5、4.0、5.5、7.0)条件下,对3个肠道有害菌(猪霍乱沙门氏菌、鸡白痢沙门氏菌、大肠杆菌K88)的抑菌圈值,统计比较抑菌性能的差异。模拟肠道pH环境的结果显示:pH 4.0、5.5、7.0条件下,BCS组和CBS组对3个指示菌均有抑制作用,且CBS组抑制效果显著大于BCS组(P<0.05);CSS组在pH 4.0条件下,对3个指示菌有抑制作用,但在pH 5.5和7.0时均无抑制作用;SDW组在pH 4.0、5.5和7.0时,对3个指示菌均无抑制作用。模拟胃pH环境的结果显示:pH 2.5条件下,4个处理组均对3个指示菌有强抑制作用,BCS、CSS、CBS 3组之间抑制效果差异不显著(P>0.05),均显著大于SDW组(P<0.05)。以上结果表明,凝结芽孢杆菌的培养液(BCS)及其离心菌悬液(CBS)可用于牛津杯抑菌试验操作,能反映菌株的抑菌性能;相反,离心上清液(CSS)的抑菌效果较弱,不适合凝结芽孢杆菌的体外抑菌试验操作。此外,本次体外抑菌研究支持了凝结芽孢杆菌在肠道环境对有害菌的有效抑制作用不受肠道pH的影响。The purpose of this study was to compare the diverse antibacterial properties of B.coagulans culture solution(BCS),or the centrifuged supernatant of B.coagulans culture solution(CSS)and the cen⁃trifuged bacterial suspension of B.coagulans cul⁃ture solution(CBS)under different pH conditions.Four treatment groups including sterile distilled water(SDW,control group),BCS,CSS,CBS were designed to determine the diameter of inhibition zone for three intestinal harmful index bacteria(S.choleraesuis,S.pullorum and E.coli K88)under the simulative gastrointestinal pH value conditions(pH 2.5,4.0,5.5 or 7.0)by the Oxford cup test in vitro.The results of simulative intestinal pH environ⁃ment showed that BCS and CBS were inhibitory against three intestinal harmful bacteria,and inhibitory efficiency of CBS was greater than that of BCS(P<0.05)under pH 4.0,5.5 and 7.0.At pH 4.0,CSS had antibacterial activity on three bacteria,but no antibacterial activity at pH 5.5 and 7.0.The SDW had no antibacterial activity on three harmful bacteria under pH 4.0,5.5 and 7.0.Under pH 2.5 of the simula⁃tive stomach condition,four groups had strong antibacterial activity on three harmful bacteria,and the in⁃hibitory efficiency of BCS,CSS and CBS were no significant difference(P>0.05),but were greater than SDW(P<0.05).In collusion,B.coagulans culture solution(BCS)and its centrifuged bacterial suspension(CBS)can be utilized for the Oxford cup test operation,which reflect the bacteriostasis of the B.coagu⁃lans strain.On the contrary,the weak inhibitory efficiency of CSS was observed,so B.coagulans centri⁃fuged supernatant was not fit to reflect the bacteriostasis of the B.coagulans strain in Oxford cup test op⁃eration.Moreover,the bacteriostasis of B.coagulans had no relevent to intestinal pH values by in vitro simulative livestock intestinal conditions.
