摘  要:【目的】研究贯叶金丝桃提取物(Hypericum perforatum extract,HPE)对急性缺氧大鼠肝肾损伤的改善作用。【方法】通过小鼠常压密闭实验模型初步筛选HPE(20%乙醇提取物、70%乙醇提取物和95%乙醇提取物)的抗缺氧活性组分。进一步建立大鼠低压低氧肝、肾损伤模型:将80只健康大鼠随机分为8组,分别为常氧空白组(NG),缺氧模型组(HG),HPE低、中、高剂量组(HPE-L、HPE-M、HPE-H,100、200、400 mg/kg),金丝桃苷低、高剂量组(HYP-L、HYP-H,50、100 mg/kg)和醋酸地塞米松组(DXM,4 mg/kg)。观察HPE对急性缺氧大鼠肝、肾组织病理学变化的影响,并测定其中氧化应激指标和相关炎症因子的变化。【结果】与模型组比较,给予HPE干预后的大鼠肝、肾组织中丙二醛(MDA)和过氧化氢(H_(2)O_(2))含量下降(P<0.01,P<0.05),谷胱甘肽(GSH)含量增加(P<0.01,P<0.05),总超氧化物歧化酶(T-SOD)和过氧化氢酶(CAT)活力有所提高(P<0.01,P<0.05)。同时,大鼠肝、肾组织中炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和血管内皮生长因子(VEGF)的释放均被抑制(P<0.01,P<0.05)。【结论】HPE对急性缺氧大鼠肝、肾损伤具有一定程度的改善作用。【Objective】This study aimed to investigate the potential ameliorative effects of Hypericum perforatum L.extract(HPE)on liver and kidney injury in acute hypoxia rats.【Method】The anti-hypoxic active components of HPE,including 20%ethanol extract,70%ethanol extract,and 95%ethanol extract,were initially screened using a normobaric hypoxia model in mice.Subsequently,a hypobaric hypoxia injury model was established using 80 rats.The rats were randomly divided into various groups:normoxia control group(NG),hypoxia model group(HG),HPE low,medium,and high dose groups(HPE-L,HPE-M,HPE-H,100,200,400 mg/kg),hyperoside low and high dose groups(HYP-L,HYP-H,50,100 mg/kg),and dexamethasone group(DXM,4 mg/kg).The effects of HPE on the histopathological changes of the liver and kidney in acute hypoxia rats were observed,and related inflammatory factors and oxidative stress indexes were measured.【Result】Compared with the model group,rats treated with HPE showed decreased levels of malondialdehyde(MDA)and hydrogen peroxide(H_(2)O_(2))(P<0.01,P<0.05),and increased levels of glutathione(GSH),total superoxide dismutase(T-SOD),and catalase(CAT)activities(P<0.01,P<0.05).Furthermore,the release of inflammatory factors interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and vascular endothelial growth factor(VEGF)was inhibited in rats treated with HPE(P<0.01,P<0.05).【Conclusion】HPE demonstrated a certain degree of ameliorative effect on liver and kidney injury in rats with acute hypoxia.
