摘  要:目的比较不同剂型和厂家葛根芩连方上市产品的化学成分差异。方法基于UV-Vis在200~600nm波长下对24批次产品进行光谱扫描,比较不同剂型样品的光谱差异;采用HPLC法,以乙腈-0.1%甲酸为流动相进行梯度洗脱,在250nm、280nm、346nm波长下同时测定各样品中葛根素等15种有效成分的含量;结合系统聚类分析(HCA)、主成分分析(PCA)、偏最小二乘判别分析(PLS-DA)对所得结果进行计量学分析。结果各剂型样品UV-Vis光谱在200~500nm范围内均有3个明显吸收峰,但峰位、峰强差异明显。在所检测的15种有效成分中,葛根素含量最高,盐酸小檗碱和黄芩苷次之;各剂型间样品含量差距大,胶囊剂有效成分总含量最高,其次为丸剂、片剂,口服液则远低于其他剂型;结合化学计量学分析表明上述测试结果可用于区分不同剂型的样品,但对不同厂家难以辨别。结论葛根芩连方不同剂型的产品之间化学成分差异大,UV-Vis和HPLC检测手段均可对其进行区分,可为该方剂的质量控制提供科学依据和参考数据。Objective To compare the chemical constituents of Gegen Qinlian formula from various dosage forms and manufactur⁃ers.Methods UV-Vis spectra of 24 batches were scanned in a wavelength range of 200~600 nm for comparison among different dosage forms.Gradient elution was carried out using acetonitrile-0.1%formic acid as mobile phase,and the contents of 15 ef⁃fective components in each sample were simultaneously determined at the wavelengths of 250nm,280nm and 346nm by HPLC method.The obtained data were analyzed by hierarchical cluster analysis(HCA),principal component analysis(PCA)and par⁃tial least squares discriminant analysis(PLS-DA).Results The UV-Vis spectrum of each dosage form showed three obvious absorption peaks in the range of 200~500nm,but with different peak position and intensity.Among the 15 active ingredients de⁃tected,puerarin was revealed as the highest content,followed by berberine chloride and baicalin.A large difference in chemical contents among different dosage forms was found,the total content of effective components in capsule was the highest,followed by pills and tablets,and oral liquid as the least.Combined with chemometrics analysis,these results can be used to distinguish sam⁃ples from different dosage forms,but it was difficult to distinguish those from different manufacturers.Conclusion The chemical constituents of Gegen Qinlian formula are quite different among different dosage forms,which can be distinguished by UV-Vis and HPLC.The present stydt can provide scientific basis and reference data for the quality control of this prescription.
