摘  要:目的 探讨远志甘草炆制前后化学成分差异及对大鼠胃肠运动的影响。方法 采用超高效液相色谱串联四极杆静电场轨道阱质谱(UPLC-Q-Exactive Orbitrap/MS)法鉴定远志与炆远志化学成分,运用正交偏最小二乘判别分析(OPLS-DA)和t检验分析远志与炆远志化学成分差异;将远志与炆远志提取物灌胃15 d后,酚红法测定大鼠胃排空率和小肠推进率,采用BL-420N+生物机能实验系统测定大鼠胃肠肌电生理活动。结果 从远志中鉴定出80个成分,从炆远志中鉴定出91个成分;远志与炆远志化学成分具有显著差异,共筛选出包括皂苷、寡糖酯、黄酮类、有机酸类、糖类等42个差异成分;远志炆制后远志皂苷、寡糖酯类成分含量呈现降低或上升趋势,[口山]酮类成分含量降低,有机酸类成分含量升高,新增甘草中皂苷类、黄酮类、香豆素类差异成分。远志可显著降低大鼠的胃排空率、小肠推进率,并可显著降低大鼠胃和十二指肠肌电快波、慢波频率,而炆远志对上述指标无明显影响。结论 远志炆制后化学成分种类和含量发生显著变化,并可有效缓解远志对大鼠胃肠运动的抑制作用。Objective To investigate differences in chemical composition and effects on gastrointestinal motility in rats of Polyga⁃lae Radix and licorice-simmered Polygalae Radix.Methods Chemical composition of Polygalae Radix and licorice-simmered Polygalae Radix was identified by ultra-high-performance liquid chromatography-Q-Exactive Orbitrap tandem mass spec⁃trometry(UPLC-Q-Exactive Orbitrap/MS),and chemical composition differences between them were analyzed by orthogonal partial least squares discriminant analysis(OPLS-DA)and t-test.After gavage of Polygalae Radix and licorice-simmered Po⁃lygalae Radix extracts for 15 days,gastric emptying rate and small intestinal propulsion rate of rats were determined by phenol red method,and gastrointestinal myophysiological activities of rats were measured by BL-420N+biological function experimental system.Results 80 components were identified from Polygalae Radix and 91 components were identified from licorice-simmered Polygalae Radix.Chemical composition of Polygalae Radix and licorice-simmered Polygalae Radix were significantly different,and a total of 42 different components including saponins,oligosaccharide esters,flavonoids,organic acids and sugars were screened out.After Polygalae Radix being simmered with licorice,contents of onjisaponins and oligosaccharide esters showed a downward or an upward trend.Content of xanthones decreased,and content of organic acids increased.Different components of saponins,fla⁃vonoids and coumarins in licorice were added.Polygalae Radix can significantly reduce gastric emptying rate and small intestinal propulsion rate of rats,and can significantly reduce rapid wave and slow wave frequencies of gastric and duodenal myoelectric in rats,while licorice-simmered Polygalae Radix has no obvious effect on the above indicators.Conclusion After Polygalae Radix being simmered with licorice,types and contents of chemical components changed significantly,and its inhibitory effect on gastroin⁃testinal motility in rats could be effectively allevi
