摘  要:目的:观察电针天枢穴对食源性肥胖小鼠胃肠运动的影响,以肠神经元标志物神经型一氧化氮合酶(nNOS)、乙酰胆碱转移酶(ChAT)等作为观察指标,探讨电针调节胃肠运动的相关肠神经机制。方法:40只C57BL/6雄性小鼠随机分为正常组(10只)、造模组(30只),采用13周高脂饲料喂养构建食源性肥胖模型,造模成功的20只小鼠随机分为模型组(10只)、电针组(10只),电针组用双极电针刺激双侧天枢穴(2 mA,2/15 Hz,20 min/次,1次/d),干预6 d后休息1 d,连续干预4周,每周称量小鼠体质量。干预结束后,检测小鼠血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)水平;检测胃排空率、小肠推进率;检测胃窦组织中神经纤维标记物蛋白基因产物9.5(PGP9.5)、nNOS、ChAT、瘦素受体(LepR)、胆囊收缩素A型受体(CCK-AR)的蛋白表达水平。结果:与模型组比较,电针组小鼠体质量、TC、HDL、LDL水平显著降低(P<0.01,P<0.05),胃排空率显著降低(P<0.01),小肠推进率有增高的趋势,PGP9.5、nNOS、LepR蛋白表达水平显著降低(P<0.05,P<0.01)。结论:电针天枢穴可改善小鼠肥胖状态,延缓胃排空,加快小肠推进,其机制可能与调控胃窦组织中PGP9.5、nNOS、LepR蛋白的表达有关。Objective:To observe the effect of electroacupuncture(EA)at Tianshu(ST 25)acupoint on gastrointestinal motility in diet-induced obesity mice,and to explore the intestinal nerve mechanism of EA regulating gastrointestinal motility with intestinal neuron markers such as neurogenic nitric oxide synthase(nNOS)and acetylcholine transferase(ChAT)as observation indicators.Methods:A total of 40 C57BL/6 male mice were randomly divided into a normal group(10 mice)and a model establishment group(30 mice,were fed a high-fat diet for 13 weeks to establish a diet-induced obesity model).Twenty mice that successfully established the model were randomly divided into a model group(10 mice)and an EA group(10 mice).The EA group was stimulated with bipolar electroacupuncture at the Tianshu(ST 25)acupoint on both sides(2 mA,2/15 Hz,20 minutes one time,once a day).After 6 days of intervention,the mice rested for 1 day and were continuously intervened for 4 weeks.The body weight of the mice was measured weekly.After the intervention,the levels of serum total cholesterol(TC),triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were measured. Gastric emptying rate and small intestine propulsion rate were detected;the protein expression levels of nerve fiber marker: protein gene product 9.5 (PGP9.5), nNOS, ChAT, leptin receptor (LepR) and cholecystokinin A receptor (CCK-AR) in gastric antrum tissue were detected. Results: Compared with the model group, the body weight, TC, HDL and LDL levels of the EA group mice were significantly reduced (P<0.01, P<0.05), the gastric emptying rate decreased (P<0.01), and the small intestine propulsion rate showed an increasing trend, the protein expression levels of PGP9.5, nNOS and LepR were significantly reduced (P<0.05, P<0.01). Conclusion: EA at Tianshu (ST 25) acupoint can improve obesity status and gastrointestinal function in mice, and its mechanism may be related to regulating the protein expression of PGP9.5, nNOS and LepR in gastric antrum.
