摘 要:本文拟对青岛胶东国际机场区域捕获的蝇类进行鉴定。实验首先进行形态学鉴定,再提取蝇类基因组DNA,利用基于线粒体COⅠ基因的DNA条形码技术进行分子鉴定,从比对结果中选取序列相似度高的5种蝇类序列构建系统进化树。结果显示,该蝇类在形态学鉴定上属于暮美根寄蝇,序列与GenBank中暮美根寄蝇(登录号:KX844373.1)COⅠ基因序列同源性为98.73%,判断此次捕获蝇类为暮美根寄蝇。查阅文献发现为山东省首次报道,故此次青岛胶东国际机场区域捕获的暮美根寄蝇为山东省新记录种。This paper aims to identify flies captured in Qingdao Jiaodong International Airport area.Firstly,the flies were identified by morphological method,then genomic DNA was extracted,and molecular identification was carried out using DNA barcoding technique based on mitochondrial CO Ⅰgene.The phylogenetic tree was constructed by selecting 5 species of flies with high sequence similarity from the comparison results.Morphological identification indicated that the flies belonged to Meigenia mutabilis,and the CO Ⅰ gene sequence of these flies was 98.73% homologous to Meigenia mutabilis in the GenBank(accession number:KX844373.1).It was the first report in Shandong province by literature review.Therefore,the Meigenia mutabilis captured at Qingdao Jiaodong International Airport area was newly recorded in Shandong Province.
