摘 要:本文优化了一种简便且实用的方法测定沉积物孔隙水中的VFAs,使用2-硝基苯肼将水样中VFAs衍生化,并利用配备紫外检测器的液相色谱在400 nm处进行检测分析。对标准曲线的线性和乙酸盐的空白进行了合理优化,标准曲线线性相关系数均高于0.999,乙酸盐的检出限由5μmol/L降低至0.5μmol/L。乳酸盐、甲酸盐和丙酸盐的检出限分别为0.2、0.3和0.3μmol/L。该方法检出限较低,精密度较高,已成功应用于胶州湾的孔隙水样品测定。Volatile fatty acids(VFAs)are the key compounds during organic matter degradation and anaerobic metabolism in marine sediments.Determination of the VFAs concentration in sedimentary pore waters can help to investigate the distribution,controlling factor and microbial activity.The existing methods for the determination of VFAs in sediment pore water have low sensitivity and high detection limit.In this study,a simple and practical method was optimized for the determination of VFAs in sediment pore water.Samples were analyzed using liquid chromatography with a UV detector at 400 nm through derivatization of VFAs with 2-nitrophenylhydrazine.The linearity of the standard curve and the blank concentration of acetate were optimized.The linear correlation coefficients of the standard curve were both higher than 0.999,and the detection limit of acetate was reduced from 5μmol/L to 0.5μmol/L.The detection limits of lactate,formate and propionate were 0.2,0.3 and 0.3μmol/L,respectively.With low detection limit and high precision,this method has been successfully applied to the determination of pore water samples in Jiaozhou Bay.
