摘　　要：目的:研究黄连总生物碱与川木香挥发油配伍对小鼠耳廓痤疮模型的影响。方法:采用二甲苯致耳肿胀法,观察黄连总生物碱与川木香挥发油不同配伍比例(1∶1、1∶2、2∶1、4∶1)的抗炎效果并筛选最佳配伍比例。采用耳廓内侧面注射痤疮丙酸杆菌(Propionibacterium acnes)建立小鼠痤疮模型,将ICR小鼠随机分为正常对照组、模型对照组、阿达帕林凝胶0.06 mg/cm^(2)组、黄连总生物碱与川木香挥发油配伍0.3、0.6、1.2 mg/cm^(2)组,每组8只。造模5 d后涂抹相应的药物,早晚各1次,连续7 d。计算各组小鼠耳廓肿胀度;胸腺、脾脏指数;观察耳廓组织病理改变;检测血清中白介素6(IL-6)、IL-1β、γ干扰素(IFN-γ)含量;免疫组化检测耳廓组织Toll样受体4(TLR4)、核转录因子-κB p65(NF-κB p65)的表达情况。结果:黄连总生物碱与川木香挥发油配伍具有一定的抗炎效果,最佳配伍比例为4∶1。与正常对照组比较,模型对照组耳廓肿胀度显著升高,胸腺和脾脏指数显著升高(P<0.01),耳廓出现明显的病理改变,血清中IL-6、IL-1β含量显著升高,IFN-γ含量显著降低(P<0.01),耳廓组织中TLR4、NF-κB p65蛋白阳性表达显著升高(P<0.01);与模型对照组相比,黄连总生物碱与川木香挥发油4∶1配伍的各剂量组耳廓肿胀度明显降低(P<0.05或P<0.01),胸腺和脾脏指数显著降低(P<0.01),耳廓病理性改变有不同程度的缓解,血清中IL-6、IL-1β含量显著降低,IFN-γ含量显著升高(P<0.01),耳廓组织中TLR4、NF-κB p65蛋白阳性表达显著降低(P<0.01)。结论:黄连总生物碱配伍川木香挥发油对小鼠痤疮有显著的治疗作用,可以有效改善痤疮的炎症反应。Objective:To study the effects of the combination of total alkaloids of Huanglian(黄连)and volatile oil of Chuanmuxiang(川木香)on auricular acne in mice.Methods:The ear swelling was induced by xylene.The anti-inflammatory effects of different ratios(1:1,1:2,2:1,and 4:1)of total alkaloids of Huanglian to volatile oil of Chuanmuxiang were observed,and the optimal ratio was screened out.A mouse model of acne was established by the injection of Propionibacterium acnes into the inner side of the ear.ICR mice were randomized into the following groups:blank control,model control,adapalene gel(0.06 mg/cm^(2)),and total alkaloids of Huanglian(黄连)combined with volatile oil of Chuanmuxiang(川木香)at doses of 0.3,0.6,and 1.2 mg/cm^(2),with 8 mice in each group.The drugs were applied 5 days after molding,once in the morning and once in the evening,for 7 days.The auricular swelling,thymus index,and spleen index were calculated,and the histopathological changes in the auricle were observed.The serum levels of interleukin(IL)-6,IL-1β,and interferon-γ(IFN-γ)were measured,and the expression of Toll-like receptor 4(TLR4)and nuclear factor-κB p65(NF-κB p65)in the auricle was measured by immunohistochemistry.Results:The combination of total alkaloids of Huanglian and volatile oil of Chuanmuxiang had an anti-inflammatory effect and the optimum ratio was 4:1.Compared with the blank control group,the model control group showed aggravated auricular swelling(P<0.01),increased thymus and spleen indexes(P<0.01),obvious pathological changes,elevated serum levels of IL-6 and IL-1βand lowered serum IFN-γlevel(P<0.01),and increased positive expression of TLR4 and NF-κB p65 in the auricular tissue(P<0.01).Compared with the model control group,the combination of total alkaloids of Huanglian and volatile oil of Chuanmuxiang at a ratio of 4:1 alleviated the auricular swelling(P<0.05 or P<0.01),decreased the thymus and spleen indexes(P<0.01),mitigated the pathological changes,lowered the serum levels of IL-6 and IL-1βand elevate
