摘 要:目的:研究肉桂挥发油化学成分组成及其体外抗氧化、抑菌和抗肿瘤等生物活性。方法:分别利用气相色谱-质谱联用(GC-MS)法和傅里叶红外光谱技术鉴定肉桂挥发油主要活性成分和官能团;紫外分光光度计测定肉桂挥发油总酚含量、DPPH自由基清除活性和β-胡萝卜素漂白抑制率;通过打孔法、微量稀释法分别测定抑菌圈直径、最小抑菌浓度(MIC)和最低杀菌浓度(MBC),考察肉桂挥发油对常见的8种致病菌和6种耐药菌的抑菌活性;MTT法测定肉桂挥发油对不同肿瘤细胞的IC50,DCFH-DA探针法检测细胞内活性氧含量。结果:肉桂挥发油共鉴定出19种化合物,总酚含量为(29.9±1.2)mg GAE/g EO,具有较高的自由基清除能力[DPPH IC50=(4.0±0.4)μg/mL],肉桂挥发油1.6 mg/mL时对β-胡萝卜素漂白的抑制率达68.6%;肉桂挥发油对8种常见细菌及6种耐药菌种均显示出一定的抑制作用;与空白对照组相比,肉桂挥发油对MDA-MB-231乳腺癌细胞显示出显著降低增殖、迁移、侵袭能力,显著诱导MDA-MB-231细胞凋亡(P<0.01)。肉桂挥发油显著上调细胞内ROS水平,上调凋亡相关蛋白Caspase-3和AIF的表达(P<0.01);引起线粒体膜电位下降,促进了细胞凋亡。结论:肉桂挥发油中成分复杂,含有多种活性成分,在抗氧化、抑菌和抗肿瘤等方面具有良好的作用。Objective:To study the chemical constituents of the essential oil of Cinnamomum cassia Presl(CEO)and its in vitro biological activities,including antioxidant,antibacterial,and anticancer properties.Methods:Gas chromatography-mass spectrometry(GC-MS)and Fouriertransform infrared spectroscopy(FTIR)were used to identify the main active constituents and functional groups of CEO.The total phenol content,DPPH free radical scavenging activity,andβ-carotene bleaching inhibition rate of CEO were measured using UV-visible spectrophotometry.The diameter of the inhibition zone,minimum inhibitory concentration(MIC),and minimum bactericidal concentration(MBC)were measured by drilling method and microdilution method.The antibacterial activity of CEO against eight common pathogens and six drug-resistant bacteria was investigated.The ICso of CEO on different tumor cells was measured using the MTT assay,and the intracellular reactive oxygen species(ROS)levels were detected using the DCFH-DA probe.Results:Nineteen compounds were identified in CEO.The total phenol content was(29.9±1.2)mg GAE/g EO.CEO exhibited high free radical scavenging activity with a DPPH ICso value of(4.0±0.4)μg/mL.At a concentration of 1.6 mg/mL,CEO inhibitedβ-carotene bleaching by 68.6%.CEO showed inhibitory effects against both the eight common bacteria and the six drug-resistant bacteria.Compared with the blank control group,CEO significantly reduced the proliferation,migration,and invasion abilities of MDA-MB-231 breast cancer cells,and induced apoptosis in MDA-MB-231 cells(P<0.01).CEO significantly increased intracellular ROS levels,upregulated the expression of apoptosis-related proteins Caspase-3 and AIF(P<0.O1),decreased mitochondrial membrane potential,and promoted cell apoptosis.Conclusion:CEO is composed of various active constituents and exhibits good effects in terms of antioxidant,antibacterial,and anticancer activities.
