摘  要:为建立适用于评价仔猪湿热泄泻证候中兽药制剂药效的动物模型,本试验应用高糖高脂+高温高湿+肠毒性大肠杆菌和高糖高脂+高温高湿+葡聚糖硫酸钠两种复合因素组合方式建立湿热泄泻大鼠模型。选用18只SD雄性大鼠随机均分为空白对照组、大肠杆菌模型组和葡聚糖硫酸钠组模型组,按不同条件分阶段造模18 d。每日监测大鼠体质量、采食饮水量、精神状态、粪便等临床表征变化,采集血液进行血常规检测,采集回肠和结肠进行组织病理变化诊断,ELISA测定血清及结肠IL-1β及IL-6表达情况,RT qPCR测定结肠IL-1β、IL-4、IL-6、TNF-α、MUC1和MUC2 mRNA表达水平。结果显示,两种模型均能造成大鼠出现不同程度的湿热泄泻临床表现和炎症反应,葡聚糖硫酸钠组肠道病理损伤较大肠杆菌组更严重;两种模型组大鼠血清IL-1β均显著升高(P<0.05),大肠杆菌组结肠IL-1β、IL-6、TNF-αmRNA水平均极显著升高(P<0.01),葡聚糖硫酸钠组结肠IL-1β、IL-6、TNF-αmRNA水平均升高(P<0.05),葡聚糖硫酸钠组MUC1 mRNA水平极显著降低(P<0.001),两种模型组结肠IL-4和MUC2 mRNA水平均极显著降低(P<0.01)。表明两种复合因素建立的湿热泄泻动物模型在临床表现、炎症反应、肠道组织病理损伤情况及结肠黏膜相关基因表达情况等多方面既有共性,亦有其自身特点,但大肠杆菌组较葡聚糖硫酸钠组造模整体表现更佳,更符合传统中兽医学湿热泄泻证候理论及生产实际要求,可为后续开展仔猪湿热泄泻相关病证研究提供科学参考。To establish an animal model suitable for evaluating the efficacy of veterinary drug preparation in piglets.A rat model of damp-heat diarrhea was established using a combination of two composite factors:high glucose+high fat+high temperature+high humidity+enterotoxic E.coli,and high glucose+high fat+high temperature+high humidity+sodium gluconate sulfate.Eighteen SD male rats were randomly divided into the blank control group,E.xoli model group and dextran sodium sulfate group,the trial lasted for 18 d.Daily clinical changes in body weight,water intake,mental status,and feces were monitored,blood was collected for routine blood tests,ileum and colon were collected for histopathological diagnosis,serum and colon IL-1β and IL-6 were detected by ELISA,and expression levels of colon IL-1β,IL-4,IL-6,TNF-α,MUC1 and MUC2 mRNA were detected by RT-qPCR.Result showed that both models caused clinical manifestations of diarrhea and inflammatory reaction in rats,the intestinal pathological damage in dextran sodium sulfate group was more serious than that the E.coli group;serum IL-1β was significantly increased in both model groups(P<0.05);IL-1β,IL-6 and TNF-α mRNA levels increased significantly in E.coli group(P<0.01);colonic IL-1β,IL-6,and TNF-α mRNA levels were increased in the sodium dextran sulfate group(P<0.05);MUC1 mRNA level was significant reduced in the dextran sulfate group(P<0.001),colonic IL-4 and MUC2 mRNA levels were significantly reduced in both model groups(P<0.01).These results showed that the damp-heat diarrhea animal model established by two composite factors have similarities in clinical manifestations,inflammatory reaction,intestinal histopathology damage and colon mucosa related gene expression,as well as their own characteristics,but the overall performance of the E.coli group was better than that of the glucan sodium sulfate group,which is more in line with the traditional veterinary theory of damp-heat diarrhea syndrome and production practical requirements,and can provide scientific reference
