摘 要:旨在探讨高铜(Cu)日粮对肉鸭肾氧化应激与自噬水平的影响。试验挑选144只1日龄北京鸭,随机分为4组:对照组、高铜Ⅰ组、高铜Ⅱ组和高铜Ⅲ组,分别饲喂铜含量为10、100、200、400 mg/kg的基础日粮。在试验的第15、30和45天采集肉鸭肾组织,检测氧化应激水平,观察超微病理学变化,分析自噬相关基因mRNA与蛋白(Beclin1、Atg5、Atg14、LC3A、LC3B、P62)的相对表达量。结果显示:与对照组相比,高铜Ⅲ组在45 d时肉鸭肾组织的T-sod、CAT活力下降,MDA和H_(2)O_(2)含量显著上升。高铜Ⅲ组肉鸭的自噬体数量较其他组增加。所有高铜处理组在30和45 d的Beclin1、Atg14、LC3A和LC3B mRNA表达水平显著升高,且45 d时高铜Ⅲ组的Beclin1和LC3B-Ⅱ/LC3B-Ⅰ的蛋白水平显著升高。高铜Ⅱ、Ⅲ组的LC3B-II荧光强度较对照组也显著增加。上述结果表明高铜诱导肉鸭肾氧化应激和自噬水平升高。To investigate the effect of high dietary copper on oxidative stress and autophagy in the kidneys of meat ducks,144 one-day-old Peking ducks were randomly divided into four groups:control group,high-copper groupⅠ,high-copper group Ⅱ and high-copper group Ⅲ,and fed with basal diets with 10,100,200 and 400 mg/kg copper content respectively.On day 15,30 and 45,the kidneys were collected,the levels of oxidative stress were detected,the ultrastructural changes were observed,and the expression levels of autophagy-related genes and proteins(Beclinl,Atg5,Atg14,LC3A,LC3B,P62)were analyzed.The results showed that compared with the control group,the activities of T-SOD and CAT decreased,and the content of MDA and H_(2)O_(2) increased significantly in the high-copper group Ⅲ on the 45th day.Additionally,more autophagosome could be observed in the ultrastructure of high-copper group Ⅲ.Moreover,mRNA levels of Beclinl,Atg14,LC3A and LC3B in the high-copper treatment group were increased significantly at 30 and45 days,and the protein levels of Beclinl and LC3B-Ⅱ/LC3B-Ⅰ in the high-copper treatment group were increased significantly at 45 days.The results of immunofluorescence showed that the fluorescence intensity of LC3B-Ⅱ in high-copper group Ⅱ and Ⅱ were markedly higher than that in the control group.These results suggest that high dietary copper can induce oxidative stress and increase autophagy in the kidneys of meat ducks.
