土家医赶气方对肝郁脾虚大鼠脾脏免疫的影响

摘　　要：目的通过研究土家医赶气方对肝郁脾虚大鼠脾脏免疫的调节作用及其机制,为临床应用提供实验依据。方法取60只雄性SD大鼠随机分为正常组、肝郁脾虚模型组、赶气方高、中、低剂量组、氟西汀组,每组10只大鼠。除正常组外的5组均以多重因素造模方法造模21 d,复制肝郁脾虚模型。完成造模后第2 d开始给药,正常组和模型组给予等量0.9%NaCl溶液,其余各组按剂量要求灌胃相应药液,1次/d,持续用药14 d。末次给药后禁食不禁水,第2 d处死大鼠,摘取各组大鼠脾脏组织,ELISA法检测炎性趋化因子配体20(CCL20)、趋化因子8(CXCL8)表达,免疫组织化学法和RT-PCR法检测核因子κB(NF-κB)、B淋巴细胞瘤-2(Bcl-2)和B淋巴细胞瘤-2同源凋亡前体蛋白(Bax)及基因表达。结果ELISA法结果显示,赶气方高、中、低剂量组CCL20、CXCL8含量明显降低,NF-κB和Bax蛋白及mRNA表达水平明显降低,Bcl-2蛋白及mRNA表达水平明显升高(P<0.05或P<0.01)。结论土家医赶气方可通过上调脾脏组织细胞免疫相关因子Bcl-2的表达,下调CCL20、CXCL8的含量及NF-κB和Bax蛋白及mRNA表达,增强脾脏细胞免疫,抑制细胞凋亡,间接达到改善肝郁脾虚症的作用。Objective To study the regulatory effect of Tujia medicine Ganqi recipe on spleen immunity in rats with liver depression and spleen deficiency and its mechanism,so as to provide experimental basis for clinical application.Method 60 male sprague-dawley rats were randomly divided into the normal group(NG),liver stagnation and spleen deficiency model group(MG),Ganqi recipe high,middle and low dose groups(GRHG,GRMG,GRLG)and fluoxetine group(FG),with 10 rats in each group.All the five groups except the normal group were modeled by multiple factors for 21 days to replicate the model of liver stagnation and spleen deficiency.The normal group and the model group were given 0.9%NaCl solution at the same dose,and the other groups were given corresponding solution by gavage once a day for 14 days.The rats were killed on the 2nd day after the last administration,and the expression of chemokines CCL20 and CXCL8 in the spleen were detected by ELISA,the protein and gene expression of NF-κb,Bcl-2 and Bax were detected by immunohistochemistry and RT-PCR.Results The results of ELISA method showed that the contents of CCL20 and CXCL8 in the high,medium and low dose groups were significantly reduced,the expression levels of NF-κB and Bax proteins and mRNA were significantly decreased,while the expression levels of Bcl-2 proteins and mRNA were significantly increased(P<0.05 or P<0.01).Conclusion Tujia medicine Ganqi recipe can enhance splenic cell immunity and inhibit cell apoptosis by up-regulating the expression of cellular immunity-related factor Bcl-2 in spleen tissues,down-regulating the content of CCL20,CXCL8,and the expression of NF-κB and Bax proteins and mRNAs,and indirectly improving the effect of liver depression and spleen deficiency.
