摘 要:脂质和葡萄糖代谢在能量稳态中发挥着重要作用,其功能紊乱可诱发代谢综合征的发生,例如肥胖和糖尿病。研究表明,参与脂质和葡萄糖代谢的骨骼属于一种内分泌器官,通过骨源性分子调节全身代谢。其中硬骨抑素(sclerostin)是一种主要由骨细胞产生的蛋白质,具有抑制骨形成的能力,其抗体已成为治疗骨质疏松症的治疗靶点。最近的证据表明硬骨抑素在脂质和葡萄糖代谢紊乱中也发挥着作用。因此,本文基于体外和体内的研究证据,综述了硬骨抑素在脂质和葡萄糖代谢的作用,以及与肥胖和糖尿病之间的关系,总结了硬骨抑素的研究进展,展望其未来在代谢性疾病治疗中的潜在作用。Lipid and glucose metabolism play crucial roles in maintaining energy homeostasis,and their dysregulation can lead to the development of metabolic disorders,such as obesity and diabetes.Studies indicate that the skeleton,involved in lipid and glucose metabolism,functions as an endocrine organ,regulating systemic metabolism through bone-derived molecules.Sclerostin is a protein mainly produced by osteocytes,possessing the ability to inhibit bone formation,and its antibodies have become therapeutic targets for treating osteoporosis.Recent evidence suggests that sclerostin also plays a role in lipid and glucose metabolism disorders.Therefore,through summarizing in vitro and in vivo researches,this article reviews the role of sclerostin in lipid and glucose metabolism,its relationship with obesity and diabetes,and potential role in the treatment of metabolic diseases in the future.
