摘  要:目的 探讨一步法用于泌尿生殖系统感染病原体核酸检测的可行性,为一步法实现广泛的临床应用提供实验基础。方法 采集2021年3月至2021年8月期间到深圳市宝安区人民医院就诊的200例泌尿生殖道感染患者分泌物,分别用一步法和传统煮沸法进行核酸提取,比较两种提取方法的结果一致性、灵敏度、抗干扰能力和重复性。结果 一步法与煮沸法的提取阳性率一致性Kappa值为0.949,一致性较高;对选取的阳性样本两种提取方法最高可检测出1∶100 000的稀释浓度;抗干扰实验干扰偏移均在10%以内;重复性实验显示两种提取方法的CV值均在5%以内。结论 两种提取方法比较差异无统计学意义,一步法操作简单快速,降低污染风险,具有非常好的临床应用价值。Objective To explore the feasibility of the one-step nucleic acid extraction method for pathogens in urogenital infections and provide an experimental basis for its wide clinical application.Methods Secretion specimens were collected from 200 patients with urogenital infection at the Peoples Hospital of Shen-zhen Baoan District from March to August 2021.Nucleic acid was extracted using both the one-step method and the conventional boiling method to compare the consistency,sensitivity,anti-interference ability and repeatabili-ty of the two extraction methods.Results The results showed that the positive consistency between the one-step method and the boiling method had a kappa value of 0.949,indicating good consistency between the two meth-ods.Both extraction methods were able to detect a dilution concentration of up to 1∶100000 for the selected posi-tive samples.The interference offsets of the two extraction methods were within 10%.Repeatability experiments demonstrated that the coefficient of variation(CV)values for both extraction methods were within 5%.Conclu-sion There was no significant difference between the two extraction methods.The one-step method is simple and quick to operate,reducing the risk of laboratory contamination and has great clinical application value.
