不同剂量三七粉对大鼠凝血功能的影响

摘　　要：目的:探讨不同剂量三七粉对大鼠凝血功能的影响。方法:选取60只健康SD大鼠,随机分为4组(对照组及三七粉低、中、高剂量组),每组15只。三七粉低、中、高剂量组分别给予三七粉1.2 g/kg,2.4 g/kg,4.8 g/kg剂量灌胃,连续给药14d。测定大鼠的凝血时间(CT)、出血时间(BT)、活化部分凝血活酶活性时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)、血浆血栓烷B(2 TXB2)和6-酮-前列腺素F1ɑ(6-keto-PGF1ɑ)水平。结果:与对照组比较,三七粉低、中剂量组CT、BT、APTT延长,TXB2减少,6-keto-PGF1ɑ增多,差异均有统计学意义(P<0.01,P<0.05)。高剂量三七粉组CT、BT、APTT缩短(P<0.05),6-keto-PGF1ɑ减少(P<0.05)。低、中、高剂量组PT、TT无明显改变。结论:低、中剂量三七粉给药对大鼠具有抗凝作用,抗凝机制与减少血栓素A(2 TXA2)和增加前列环素(PGI2)的释放以及促进内源性凝血过程有关,而大剂量三七粉对大鼠有促凝作用,与加强内源性凝血过程和抑制PGI2释放有关。Objective:To explore the effect of different doses of Sanqi powder on blood coagulation in rats.Methods:60 healthy SD rats were randomly divided into 4 groups(control group,low-dose Sanqi powder group,medium-dose Sanqi powder group and high-dose Sanqi powder group),with 15 rats in each group.The rats in the low-dose Sanqi powder group,medium-dose Sanqi powder group and high-dose Sanqi powder group were given Sanqi powder 1.2 g/kg,2.4 g/kg and 4.8 g/kg by gavage,respectively,for 14 consecutive days.The clotting time(CT),bleeding time(BT),activated partial thromboplastin time(APTT),prothrombin time(PT),thrombin time(TT),plasma thromboxane B2(TXB2)and 6-keto-prostaglandin F1ɑ(6-keto-PGF1ɑ)were measured.Results:Compared with those in the control group,CT,BT,APTT were prolonged,TXB2 was decreased,6-keto-PGF1ɑwas increased in the low-and medium-dose Sanqi powder groups(P<0.01,P<0.05).In high-dose Sanqi powder group,CT,BT and APTT were shortened(P<0.05),and 6-keto-PGF1ɑdecreased.The differences were all statistically significant(P<0.05).In high-dose Sanqig powder group,CT,BT and APTT were shortened(P<0.05),and 6-keto-PGF1ɑdecreased(P<0.05).There were no significant changes in PT and TT in low,medium and high dose groups.Conclusion:Low and medium doses of Sanqi powder have anticoagulant effect on rats.The anticoagulant mechanism is related to reducing thromboxane A2(TXA2),increasing the release of prostacyclin(PGI2)and promoting endogenous coagulation process,while large doses of Sanqi powder have procoagulant effect on rats,which is related to enhancing endogenous coagulation process and inhibiting the release of PGI2.
