摘　　要：目的筛选小鼠触须毛囊外根鞘(ORS)在体外培养再生的最适双氢睾酮(DHT)浓度,通过检测Dermo-1、CD11b探索其对毛囊外根鞘再生的影响。方法利用冰冻切片技术HE染色观察毛囊外根鞘在不同DHT浓度培养条件下的再生程度,利用免疫荧光技术鉴定Dermo-1、CD11b在最适DHT浓度条件下在毛囊外根鞘中的表达情况。结果浓度为10^(-5) mol/L、10^(-6) mol/L的DHT对毛囊外根鞘体外再生起抑制作用,但10^(-9) mol/L的DHT促进其体外再生。并且Dermo-1、CD11b在毛囊外根鞘再生过程中均有表达。结论毛囊外根鞘在体外培养第5天时再生的结构最完整,10^(-9) mol/L的DHT与体内浓度最为接近,毛囊干细胞对创伤修复和组织结构再生都发挥着重要作用。Objective To screen the optimal concentration of dihydrotestosterone(DHT)for the regeneration of the outer root sheath(ORS)of mouse hair follicle in vitro,and the effects of DHT on the regeneration of ORS of hair follicle were investigated by detecting Dermo-1 and CD11b.Methods The regeneration degree of hair follicle ORS under different DHT concentrations was observed by frozen sections and HE staining,and the expression of Dermo-1 and CD11b in hair follicle ORS was identified by immunoflurescence under the optimal DHT concentrations.Results The results showed that DHT concentrations at 10^(-5) mol/L and 10^(-6) mol/L had inhibitory effect on regeneration of hair follicle ORS,while DHT at 10^(-9) mol/L promoted them regeneration in vitro,and both of Dermo-1 and CD11b expressed positivly during hair follicle ORS regeneration as well.Conclusion The regenerated structure of hair follicle ORS is the most complete on Day 5 culture in vitro,and the concentration of DHT at 10^(-9) mol/L is the closest to that in vivo.Hair follicle stem cells play an important role in wound healing and tissue regeneration.
