摘 要:目的:探讨白藜芦醇对糖尿病心脏微血管损伤的作用及机制。方法:分离、培养大鼠心脏微血管内皮细胞,随机分为正常组、甘露醇组、高糖组、高糖+白藜芦醇组及高糖+白藜芦醇+compound C组。采用流式细胞仪检测细胞活性氧水平,试剂盒法检测超氧化物歧化酶活性和丙二醛含量,免疫印迹法检测腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)的活性。结果:与正常组相比,高糖组的活性氧水平和丙二醛含量增高,超氧化物歧化酶活性和p-AMPK/AMPK值降低;与高糖组相比,高糖+白藜芦醇组的活性氧水平和丙二醛含量降低,超氧化物歧化酶活性和p-AMPK/AMPK值升高;与高糖+白藜芦醇组相比,高糖+白藜芦醇+compound C(AMPK抑制剂)组中活性氧水平和丙二醛含量升高,超氧化物歧化酶活性和p-AMPK/AMPK值降低,差异有统计学意义(均P<0.05)。结论:白藜芦醇通过激活AMPK减轻糖尿病心脏微血管损伤。Objective:To investigate the effect and mechanism of resveratrol on diabetic cardiac microvascular injury.Methods:Rat cardiac microvascular endothelial cells were isolated and cultured,and randomly divided into normal glucose group,mannitol group,high glucose group,high glucose+resveratrol group and high glucose+resveratrol+compound C group.The cellular reactive oxygen species level was detected by flow cytometry,superoxide dismutase activity and malondialdehyde content were detected by commercial assay kit,and the activity of AMP-activated protein kinase(AMPK)was detected by Western blot.Results:Compared with the normal group,reactive oxygen species level and malondialdehyde content were increased,and superoxide dismutase activity and p-AMPK/AMPK value were decreased in the high glucose group.Compared with the high glucose group,reactive oxygen species level and malondialdehyde content were decreased,and superoxide dismutase activity and p-AMPK/AMPK value were increased in the high glucose+resveratrol group.Compared with the high glucose+resveratrol group,reactive oxygen species levels and malondialdehyde content were increased and superoxide dismutase activity and p-AMPK/AMPK values were decreased in the high glucose+resveratrol+compound C(AMPK inhibitor)group,and the differences were statistically significant(all P<0.05).Conclusion:Resveratrol attenuates diabetic cardiac microvascular injury by activating AMPK.
