TLR4/NF-κB p65高表达对皮肤鳞状细胞癌增殖、侵袭和转移的影响

摘　　要：目的探讨与分析Toll样受体4(Toll-like recepTor 4,TLR4)/核因子KappaB(NF-κB)p65高表达对皮肤鳞状细胞癌增殖、侵袭和转移的影响。方法对数生长期的人皮肤鳞状细胞癌A431细胞株分为三组,空白组、对照组与实验组,分别转染DMEM培养基、pc3.0空载体、pc3.0-TLR4NF-κB p65载体,检测皮肤鳞状细胞癌增殖、侵袭和转移表达情况。结果转染24 h、48 h后,实验组的TLR4、NF-κB p65 RNA表达水平显著高于空白组与对照组,差异有统计学意义(P<0.05),空白组与对照组比较,差异无统计学意义(P>0.05)。实验组的细胞增殖指数显著低于空白组与对照组,差异有统计学意义(P<0.05),对照组与空白组比较,差异无统计学意义(P>0.05)。实验组的细胞侵袭指数与细胞转移指数都明显低于空白组与对照组,差异有统计学意义(P<0.05),对照组与空白组比较,差异无统计学意义(P>0.05)。实验组的Caspase-9/Myc蛋白表达水平显著高于空白组与对照组,差异有统计学意义(P<0.05),空白组与对照组比较,差异无统计学意义(P>0.05)。结论TLR4/NF-κB p65高表达能抑制皮肤鳞状细胞癌细胞的增殖、转移与侵袭,也可促进Caspase-9/Myc蛋白的表达,从而发挥促进肿瘤细胞凋亡的作用。Objective To explore and analysis the effects of Toll like receptor 4(TLR4)/nuclear factor NF-κBp65 high expression on the proliferation,invasion,and metastasis of skin squamous cell carcinoma.Methods Human skin squamous cell carcinoma A431 cell line with logarithmic growth phase were divided into three groups-blank group,control group,and experimental group.They were transfected with DMEM medium,pc3.0 empty vector,and pc3.0-NF-κBp65,respectively vector.All the cells were used to detect the expression of proliferation,invasion,and metastasis in skin squamous cell carcinoma.Results After 24 and 48 hours of transfection,the expression of TLR4 and NF-κBp65 in the experimental group were signifi cantly higher than that of the blank group and control group(P<0.05),and there were no signifi cant diff erence compared between the blank group and control group(P>0.05).The cell proliferation index of the experimental group were signifi cantly lower than that of the blank group and control group(P<0.05),and there were no signifi cant diff erence compared between the control group and the blank group(P>0.05).The cell invasion index and cell metastasis index of the experimental group were significantly lower than those of the blank group and control group(P<0.05),while there were no significant diff erence compared between the control group and the blank group(P>0.05).The expression level of Caspase-9/Myc protein in the experimental group were signifi cantly higher that of the blank group and control group(P<0.05),and there were no significant difference compared between the blank group and control group(P>0.05).Conclusion TLR4/NF-κBp65 high expression can inhibit the proliferation,metastasis,and invasion of skin squamous cell carcinoma cells,as well as promote the expression of Caspase-9/Myc protein,thereby exerting promote apoptotic eff ect on tumor cells.
