摘  要:为了揭示三角梅褪绿脉带病毒(Bougainvillea Chlorotic Vein Banding Virus, BCVBV)在广州发生情况及其系统发育关系,本研究从广东省广州市采集疑似三角梅病样42份,经PCR扩增和序列分析,共获得9个BCVBV广州分离物.对这9个分离物及GenBank中相应分离物进行序列分析,并构建系统发育树.结果表明,BCVBV分离物可划分为2个亚组,BCVBV广州分离物与中国台湾、海南分离物属于同一亚组,亲缘关系较近;巴西分离物属于另一亚组,与广州分离物亲缘关系较远.BCVBV序列同源率分析表明,9个广州分离物基因序列核苷酸和氨基酸序列同源率分别为99%~100%和98.1%~100%,与其他分离物核苷酸和氨基酸序列同源率分别为79.1%~98.1%和89.8%~100%.本研究揭示了BCVBV广州分离物与其他分离物之间的系统发育关系,为三角梅BCVBV的研究和三角梅新品种培育奠定了基础.In order to reveal the occurrence and phylogenetic relationships of Bougainvillea chlorotic vein banding virus(BCVBV)in Guangzhou,42 suspected disease samples were collected from Guangzhou,Guangdong Province,and a total of 9 BCVBV Guangzhou isolates were identified by PCR.The corresponding sequences of 9 Guangzhou isolates and BCVBV isolates in GenBank were analyzed,and a phylogenetic tree was constructed based on 9 BCVBV Guangzhou isolates and other BCVBV isolates in GenBank.The phylogenetic analysis showed that all BCVBV isolates were classified into two subgroups,BCVBV Guangzhou isolates were clustered in the same branch,and shared a closer phylogenetic relationship with the isolates from Taiwan and Hainan.However,the Brazil isolates were grouped in a different branch,which had more distantly related to the BCVBV Guangzhou isolates.The identities of nucleotide and amino acid sequence of 9 BCVBV Guangzhou isolates shared 99%~100%and 98.1%~100%,respectively,while that with other isolates from GenBank showed 79.1%~98.1%and 89.8%~100%,respectively.This study reveals the evolutionary relationship between BCVBV Guangzhou isolates and other isolates,laying the foundation for the study of BCVBV and the breeding of Bougainvillea spectabilis.
