摘 要:为了确认菊花粕在饲料中的添加对高效液相色谱法检测饲料中的金霉素具有干扰作用,本研究采用饲料中金霉素测定(GB/T 19684-2005)高效液相色谱法,以来源于不同饲料生产厂家配方中含有菊花粕的配合饲料2批,菊花粕原料3批,配方中不含有菊花粕的配合饲料3批为研究对象。验证金霉素阳性添加样品的回收率,不同样品色谱峰保留时间、不同稀释倍数与含量之间的关系等。由研究结果看出:金霉素标准品保留时间为7.193 min,金霉素阳性添加回收率在95.9%~104.2%之间;不含菊花粕的样品及空白样品在金霉素的保留时间未出现色谱峰,含菊花粕的样品及菊花粕原料该时间均有色谱峰,峰形良好,根据保留时间被认定为金霉素,且保留时间允许误差不超过2%;该色谱峰所代表的未知物质的含量与稀释倍数呈负相关,研究测得菊花粕原料中该未知物含量在97.8~105.4 g·kg^(-1)之间。因此,本研究结果表明:利用高效液相色谱法(GB/T 19684-2005)测定饲料中金霉素时,饲料配方中如果含有菊花粕,会对金霉素的检测具有干扰作用。In order to confirm that the addition of chrysanthemum meal in feed can interfere with the detection of chlorotetracycline in feed by high performance liquid chromatography(HPLC).In this study,the determination of chlorotetracycline in feed(GB/T 19684-2005)was used by HPLC.Two batches of complex feed containing chrysanthemum meal,three batches of raw materials of chrysanthemum meal and three batches of complex feed without chrysanthemum meal in the formula of different feed manufacturers were studied.To verify the recovery rate of chlorotetracycline positive added samples,the relationship among chromatographic peak retention time of different samples,different dilution times and contents,etc.The results showed that the retention time of chlorotetracycline standard was 7.193 min,and the recovery rate of chlorotetracycline positive addition was 95.9%-104.2%.The samples without chrysanthemum meal and blank samples did not have chromatographic peaks at the retention time of chlorotetracycline,the samples containing chrysanthemum meal and chrysanthemum meal raw materials had chromatographic peaks at the time,and the peak shape was good,and it was identified as chlorotetracycline according to the retention time,and the allowable error of retention time was not more than 2%.The content of unknown substances represented by the chromatographic peak was negatively correlated with dilution times,and the content of unknown substances measured in chrysanthemum meal raw materials was between 97.8 g·kg^(-1)and 105.4 g·kg^(-1).Therefore,the results of this study showed that the determination of chlorotetracycline in feed by high performance liquid chromatography(GB/T 19684-2005)would interfere with the detection of chlorotetra-cycline if the feed formula contained chrysanthemum meal.
