蒙药材东北齿缘草的质量标准研究

摘　　要：目的建立东北齿缘草药材的质量标准,为东北齿缘草质量控制和评价提供参考。方法参照2020年版《中华人民共和国药典》四部通则相关方法,对东北齿缘草的水分、总灰分、酸不溶性灰分和浸出物进行测定。用高效液相色谱法,以Hypersil ODS(300 mm×4.6 mm,2.5μm)为色谱柱,以4 mL·L^(−1)磷酸水-乙腈为流动相,梯度洗脱,流速为1.0 mL·min^(−1),柱温为35℃,检测波长为360 nm,建立东北齿缘草中绿原酸、芦丁和异槲皮苷的定量分析方法。结果东北齿缘草中水分、总灰分和酸不溶性灰分不得超过10.0%、7.0%、2.0%,浸出物不得低于20%。在HPLC条件下,绿原酸、芦丁和异槲皮苷质量浓度分别在1.42~4.25、0.80~2.40、0.14~0.42μg·mL^(−1)范围内呈良好的线性关系,平均回收率分别为96.33%、95.50%、95.63%,RSD值分别为1.7%、0.9%、1.4%。结论建立的东北齿缘草定量方法重复性好、灵敏度高、专属性强,可用于蒙药材东北齿缘草的质量控制。Objective To establish the quality standard of Eritrichium mandshuricum M.Pop.and provide reference for quality control and evaluation of E.mandshuricum M.Pop..Methods The moisture,total ash,acid-insoluble ash and extracts of E.mandshuricum M.Pop.were determined by referring to the relevant methods compiled in Volume 4,General Chapters in the Chinese Pharmaco⁃poeia(2020 edition).A method for the quantitative analysis of chlorogenic acid,rutin and isoquercitrin in E.mandshuricum M.Pop.was established by high performance liquid chromatography(HPLC).The separation was performed by gradient elution,using Hypersil ODS(300 mm×4.6 mm,2.5μm)as the column at 35℃,at a flow rate of 1.0 mL·min^(−1),with 4 mL·L^(−1) phosphoric acidacetonitrile as the mobile phase,with a detection wavelength at 360 nm.Results The moisture content,total ash content and acid insoluble ash content of E.mandshuricum M.Pop.should not exceed 10.0%,7.0%and 2.0%,respectively,and the extract should not be less than 20%.The mass concentration of chlorogenic acid,rutin and isoquercitrin showed good linear relationship in the range of 1.42~4.25μg·mL^(−1),0.80~2.40μg·mL^(−1)and 0.14~0.42μg·mL^(−1),respectively.The average recovery rates were 96.33%,95.50%and 95.63%,and RSD were 1.7%,0.9%and 1.4%,respectively.Conclusion The established quantitative method has good repeatability,high sensitivity and specificity,and can be used for the quality control of Mongolian medicinal herb E.mandshuricum M.Pop..
