摘  要:本研究旨在基于CaMKⅡ-NLRP3通路探究丹七片改善心功能、减轻心肌纤维化的药效机制。采用左冠状动脉前降支结扎术制备小鼠心肌纤维化模型,利用BATMAN数据库和分子对接分析、苏木精-伊红染色、马松染色、免疫荧光染色、蛋白免疫印迹等技术评价小鼠心功能、病理改变和通路蛋白表达情况。结果显示,丹七片治疗后小鼠心功能明显改善、胶原沉积减少、纤维化程度减轻,钙/钙调蛋白依赖性蛋白激酶Ⅱ(Calcium/calmodulin-dependent protein kinase typeⅡ,CaMKⅡ)活化以及NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor protein 3,NLRP3)通路的蛋白质表达明显减少。结果表明,丹七片通过抑制CaMKⅡ-NLRP3通路改善心功能,减轻心肌纤维化。The aim of this study is to explore the mechanism of Danqi pills in improving cardiac function and reducing myocardial fibrosis based on the CaMKⅡ-NLRP3 pathway.A mouse myocardial fibrosis model was prepared by left anterior descending branch ligation.Batman database and molecular docking analyzing,hematoxylin-eosin staining,Masson staining,immunofluorescent staining and Western blot were used to evaluate the cardiac function,pathological changes and pathway protein expression of mice.The results showed that after Danqi pills treatment,the cardiac function of mice was significantly improved,collagen deposition was reduced,the degree of fibrosis was alleviated,the activation of calcium/calmodulin-dependent protein kinaseⅡ(CaMKⅡ),and the expression of NOD-like receptor pyrin domain-containing protein 3(NLRP3)related pathway proteins were significantly decreased.The results indicate that Danqi pills can improve cardiac function and reduce myocardial fibrosis by inhibiting the CaMKⅡ-NLRP3 pathway.
