摘 要:目的鳞状细胞癌(squamous cell cancer,SCC)是发生于表皮或附属器细胞的一种恶性肿瘤,它的高异质性是个性化治疗的主要挑战。从分子层面对SCC的研究已经得到了很大的进展,但是对SCC的代谢亚类研究仍然需要更多的探索。为了找到SCC潜在的代谢亚类,本文提出了一个SCC代谢亚类识别及特征分析的流程。方法基于癌症基因组图谱(The Cancer Genome Atlas,TCGA)的1316名SCC患者的代谢基因表达谱,使用共识聚类确定了两个亚类,分析了亚类的代谢特征、临床特征、免疫浸润以及药物敏感性,并构建了一个用于预测鳞状细胞癌亚类的分类模型。结果C1亚类具有较高的代谢活性、较低的免疫浸润和良好的预后。C2亚类具有较低的代谢活性、免疫检查点的高表达和较差的预后。结论本文基于代谢基因的表达谱确定了新的SCC分类,并为SCC的诊断及个性化治疗提供了有价值的信息。Objective Squamous cell cancer(SCC)is malignant tumor that occurs in epidermal or adnexal cells,and their high heterogeneity is a major challenge for personalized therapy.Much progress has been made in the study of SCC at the molecular level,but more exploration of the metabolic subclasses of SCC is still needed.In order to find the potential metabolic subclasses of SCC,a process for the identification and characterization of metabolic subclasses of SCC is proposed in this paper.Methods Based on the analysis of metabolic gene expression profiles of 1316 SCC patients from The Cancer Genome Atlas(TCGA),two subclasses were identified by consensus clustering.The correlative metabolic characteristics,clinical characteristics,immune infiltration and drug sensitivity were studied.And a classification model for predicting the subtypes of squamous cell carcinoma was constructed.Results Subclass C1 had higher metabolic activity,lower immune infiltration and favorable prognosis.The C2 subclass had lower metabolic activity,high expression of immune checkpoints,and poor prognosis.Conclusions This study establishes a new SCC classification based on the gene expression profiles of metabolic genes and provides valuable information for the diagnosis and personalized treatment of SCC.
