摘 要:近年来,山东省部分地区谷子线虫病频繁发生、危害严重。为明确山东省谷子线虫病的病原种类,2021—2022年从山东省收集谷穗样本9个,采用组织破碎-贝曼漏斗法分离获得线虫,利用形态学和分子生物学方法鉴定线虫种类。结果发现,从临沂、济宁、聊城、济南和潍坊的6个谷穗样本中分离到植物线虫,经形态学鉴定为同一物种,并对其为害症状和雌、雄虫主要形态特征进行了描述。同时结合基于rDNA-ITS与28S D2/D3区序列构建系统发育树和贝西滑刃线虫(Aphelenchoides besseyi)特异性引物PCR扩增等生物学鉴定方法,最终将为害线虫鉴定为贝西滑刃线虫。鉴定结果为山东省谷子线虫病的发生、分布和危害状况的研究提供数据参考。In recent years,millet nematode disease has occurred frequently and seriously in some areas ofShandong Province.To clarify the pathogenic species of millet nematode disease in Shandong Province,nine milletearhead samples were collected from Shandong Province in 2021-2022,and then the nematodes were isolated andobtained by tissue crushing-Behman funnel method,and identified by morphological and molecular biological methods.The results showed that nematodes could be extracted from six millet earhead samples from Linyi,Jining,Liaocheng,Jinan and Weifang cities.Through morphological identification,the six nematode populations were foundto be the same species,and then the harm symptom and main morphological characteristics of female and malenematodes were described.Combined with the phylogenetic tree constructed based on rDNA-ITS and 28S D2/D3 region sequences and PCR amplification with GU-F/GU-R primers specific for Aphelenchoides besseyi,the nematodepathogen was identified as A.besseyi.These results could provide references for researches on occurrence,distribution and harm status of millet nematode disease in Shandong Province.
