摘 要:目的:探讨人脐带间充质干细胞(hUCMSCs)移植对围绝经期大鼠卵巢功能改善情况。方法:选取13~15月龄雌性大鼠30只,构建围绝经期大鼠模型。将围绝经期鼠分为hUCMSCs移植组和空白组,每组15只,另选15只健康雌性大鼠为青年对照组。hUCMSCs移植前,检测大鼠血清E 2、AMH、FSH水平;hUCMSCs移植后2、3、4周,每组随机选取5只大鼠检测血清E 2、AMH、FSH,并取卵巢组织检测Bcl-2、Bax、Caspase-3蛋白表达水平。结果:hUCMSCs移植后,hUCMSCs组大鼠E 2、AMH高于空白组,FSH低于空白组,差异有统计学意义(P<0.05);hUCMSCs组大鼠卵巢始基卵泡、初级卵泡、次级卵泡、窦状卵泡数均多于空白组,差异有统计学意义(P<0.05);移植后4周hUCMSCs组大鼠卵巢组织中Bcl-2、Bcl-2/Bax高于空白组,Bax、Caspase-3低于空白组,差异有统计学意义(P<0.05)。结论:hUCMSCs移植后大鼠E 2、AMH升高,FSH降低;卵巢组织中Bcl-2、Bcl-2/Bax比率提高,Bax、Caspase-3下降,进一步支持了hUCMSCs在调节卵巢细胞凋亡过程中可能发挥作用。Objective:To explore the improvement of ovarian function in perimenopausal rats after transplantation of human umbilical cord mesenchymal stem cells(hUCMSCs).Methods:Thirty female rats aged 13~15 months were selected to construct a perimenopausal rat model.Perimenopausal mice were divided into hUCMSCs transplantation group and blank group,with 15 mice in each group.Another 15 healthy female rats were selected as the young control group.Before hUCMSCs transplantation,venous blood was collected from rats in each group to detect serum E 2,AMH,and FSH levels.2 weeks,3 weeks,and 4 weeks after hUCMSCs transplantation,5 rats from each group were randomly selected to detect serum E 2,AMH,and FSH,and ovarian tissue was collected to detect Bcl-2.Bax and Caspase-3 protein expression levels.Results:After hUCMSCs transplantation,the E 2 and AMH of the rats in the hUCMSCs group were higher than those of the blank group,and the FSH was lower than that of the blank group,and the differences were statistically significant(P<0.05).The ovarian primordial follicles,primary follicles,and secondary follicles of rats in the hUCMSCs group,the number of antral follicles in the hUCMSCs group was higher than that in the blank group,and the difference was statistically significant(P<0.05).4 weeks after transplantation,the expressions of Bcl-2 and Bcl-2/Bax in the hUCMSCs group were higher than those in the blank group,and the expressions of Bax and Caspase-3 were lower than those in the blank group,the difference was statistically significant(P<0.05).Conclusion:After the transplantation of hUCMSCs,levels of E 2 and AMH in rats increased while FSH decreased.Additionally,the increase in ovarian Bcl-2 and Bcl-2/Bax ratio as well as the decrease in Bax and Caspase-3 further supports the potential role of hUCMSCs in regulating the process of ovarian cell apoptosis.
