中药溃疡油对大鼠化疗外渗性损伤创面组织VEGF、EGF、bFGF表达的影响

摘　　要：目的:探究中药溃疡油促进大鼠化疗外渗性损伤创面的愈合机制。方法:将80只吡柔比星外渗性皮肤损伤模型大鼠随机分成实验组和对照组各40只。实验组采用中药溃疡油涂抹,对照组用0.9%氯化钠注射液涂抹,每日外敷给药2次,化疗外渗性皮肤损伤敷药后第2、6、10、18、24天各处死大鼠8只,分别以放射免疫法测量外渗性皮炎组织中血管内皮生长因子(VEGF)、表皮生长因子(EGF)、碱性成纤维细胞生长因子(bFGF)的水平。结果:实验组大鼠的外渗性损伤创面组织中VEGF、EGF、bFGF水平在敷药后第2、6、10、18、24天均显著高于对照组,差异均具有统计学意义(P<0.01)。结论:中药溃疡油可显著减轻外渗性创面的损伤,减少微血管渗透性、减轻血管炎症反应和组织水肿的作用可能是通过增加创面组织VEGF、EGF、bFGF的表达来实现,推测可能是中药溃疡油治疗化疗外渗性损伤有效的重要机制之一。Objective To explore the traditional Chinese medicine ulcer oil in rats induced by chemotherapy extravasation injury.Methods A total of 80 rats with skin injury induced by extravasation of pirarubicin were randomly divided into the experimental group and the control group with 40 rats each.The experimental group applied a traditional Chinese medicine ulcer oil,while the control group used 0.9%sodium chloride injection,which was externally applied twice a day.Rats in the chemotherapy group were sacrificed on days 2,6,10,18 and 24 after drug application for extravasation-induced skin injury.The levels of vascular endothelial growth factor(VEGF),epidermal growth factor(EGF)and basic fibroblast growth factor(bFGF)in exosmotic dermatitis were measured by radioimmunoassay.Results In the rats of the experimental group,the levels of VEGF,EGF,and bFGF in the tissue of extravasation-induced wounds were significantly higher than those in the control group at 2,6,10,18 and 24 days after treatment,with statistically significant differences(P<0.01).Conclusion The traditional Chinese medicine ulcer oil significantly alleviated the damage of extravasation-induced wounds,reduced microvascular permeability,and attenuated vascular inflammatory response and tissue edema,which may be achieved by increasing the expression of VEGF,EGF and bFGF in wound tissue.It is speculated that this may be one of the important mechanisms by which the traditional Chinese medicine ulcer oil is effective in treating extravasationinduced injuries.
