摘  要:目的:运用网络药理学预测贯叶金丝桃有效成分治疗轻中度抑郁症的主要作用靶点及相关信号通路。方法:运用中药系统药理学数据库与分析平台(TCMSP)获取贯叶金丝桃的有效成分及主要作用靶点,通过软件Cytoscape 3.9.1构建药物-有效成分-疾病靶点的网络关系并进行交联分析,通过STRING数据库绘制蛋白质-蛋白质相互作用网络图,用R软件、Perl及相关运行脚本筛选出贯叶金丝桃治疗轻中度抑郁症的有效成分及主要作用靶点,并生成基因本体论(GO)功能富集分析图和京都基因与基因组百科全书(KEGG)通路富集分析图。结果:共预测得到贯叶金丝桃治疗轻中度抑郁症的6种有效成分和7个有效作用靶点,分别为槲皮素、山柰酚、木犀草素、(+)-儿茶素、表儿茶素、谷甾醇;信号转导因子和转录激活因子(Signal Transducer and Activator of Transcription,STAT)1、FOS原癌基因(Fos Proto-Oncogene,FOS)、周期蛋白依赖激酶抑制因子1(Cyclin Dependent Kinase Inhibitor 1,CDKN1)、周期蛋白D1(Cyclin D1,CCND1)、丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、RELA癌基因(RELA Proto-Oncogene,NF-KB Subunit,RELA)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)。并通过通路富集分析推测贯叶金丝桃对轻中度抑郁症的作用机制可能和磷脂酰肌醇3激酶(Phosphatidylinositol 3-kinase,PI3K)-蛋白激酶B(Akt)信号通路、晚期糖基化终末产物(Advanced Glycation End Products,AGE)-晚期糖基化终末产物受体(Advanced Glycation End Product Receptor,RAGE)信号通路、化学致癌活性氧(Reactive Oxygen Species,ROS)、肿瘤坏死因子、细胞凋亡等有关,并发现STAT1、核因子-κB(Nuclear Factor-κB,NF-κB)、EGFR、CDKN1(P21)、AKT1、CCND1、FOS等靶点基因可能在轻中度抑郁症中起到关键作用。结论:通过网络药理学分析并推测了贯叶金丝桃可能通过多成分、多基因、多靶点的方式治疗轻中度抑�Objective:To predict the main targets and related signaling pathways of Guanye Jinsitao(Hypericum perforatum) in the treatment of mild to moderate depression by network pharmacology.Methods:The effective components and main targets of Guanye Jinsitao were obtained TCMSP,and the network relationship of drugs-active components-disease targets was constructed using software Cytoscape 3.9.1 for cross-linking analysis,and the PPI network diagram was drawn in the STRING database.The effective components and main targets of Guanye Jinsitao in the treatment of mild and moderate depression were screened using R software,Perl and related running scripts,and the GO functional enrichment analysis diagram and KEGG pathway enrichment analysis diagram were generated.Results:Six effective components and seven effective targets of Guanye Jinsitao in the treatment of mild and moderate depression were predicted,which were quercetin,kaempferol,luteolin,(+)-catechin,epicatechin and sitosterol,and STAT1,FOS,CDKN1,CCND1,AKT1,RELA,EGFR respective.And through pathway enrichment analysis,it was speculated that the mechanism of Guanye Jinsitao on mild to moderate depression might be related to PI3K-Akt signaling pathway,AGE-RAGE signaling pathway,chemically carcinogenic ROS,tumor necrosis factors,apoptosis,etc.It was found that STAT1,NF-κB,EGFR,CDKN1(P21),AKT1,CCND1,FOS and other target genes may play a key role in mild to moderate depression.Conclusion:The effects and characteristics of Guanye Jinsitao in the treatment of mild to moderate depression by means of multicomponent,multi-gene and multi-target are speculated through network pharmacological analysis,which provides a theoretical basis and a new starting point for further research and development of antidepressant drugs.
