摘 要:目的对黑老虎Kadsura coccinea果实中的化学成分及抗炎活性进行研究。方法通过硅胶、ODS、Sephadex LH20、MCI等多种柱色谱技术进行分离、纯化,根据波谱数据和理化性质鉴定化合物的结构。构建脂多糖(lipopolysaccharide,LPS)诱导小鼠巨噬细胞RAW264.7释放NO的模型评价化合物的抗炎活性。结果从黑老虎果实75%乙醇提取物中分离得到11个化合物,分别鉴定为(aS,7R,8R,8'S,7'R)-7-苄酰氧基-7,7',8,8'-四氢-4,5-亚甲二氧基-3,3',4',5',7'-五甲氧基-8,8'-二甲基-二苯并[a,c]环辛烯(1)、3,4-二羟基-3',4'-二甲氧基-6,7'-环木脂素(2)、kadsuralignan H(3)、(+)-(8S,8'R)-4,4'-二羟基-3,3',5'-三甲氧基木脂烷(4)、前戈米辛(5)、松脂醇(6)、aromadendrane-4β,10β-diol(7)、nephthediol(8)、oplodiol(9)、1β,11-dihydroxy5-eudesmene(10)、pubinernoid A(11)。结论化合物1为1个新的联苯环辛烯型木脂素类化合物,命名为血藤果素A;化合物2、4、7~11为首次从该属中分离得到。在50μmol/L的浓度下,化合物4可以有效的抑制LPS诱导RAW264.7细胞NO的释放,抑制率为64.45%。Objective To study the chemical constituents and anti-inflammatory activity in the fruits of Heilaohu[Kadsura coccinea(Lem.)A.C.Smith].Methods The chemical constituents from fruits of K.coccinea were isolated and purified by silica gel,ODS,Sephadex LH-20,and MCI column chromatography.Their structures were determined by analysis of spectroscopic data and physicochemical properties,and by comparison with the reported data.All compounds were evaluated for their anti-inflammatory activity in lipopolysaccharide(LPS)-stimulated RAW264.7 macrophage cells.Results A total of eleven compounds were isolated from 75%ethanol extract of the fruits of K.coccinea,and they were identified as(aS,7R,8R,8'S,7'R)-7-dibenzoyloxy-7,7',8,8'-tetrahydro-4,5-methylenedioxy-3,3',4',5',7'-pentamethoxy-8,8'-dimethyl-dibenzo[a,c]cycloocten(1),3,4-dihydroxy-3',4'-dimethoxy6,7'-cyclolignan(2),kadsuralignan H(3),(+)-(8S,8'R)-4,4'-dihydroxy-3,3',5'-trimethoxylignan(4),pregomisin(5),pinoresinol(6),aromadendrane-4β,10β-diol(7),nephthediol(8),oplodiol(9),1β,11-dihydroxy-5-eudesmene(10),pubinernoid A(11),respectively.Conclusion Compound 1 is a new dibenzocyclooctadiene lignan named kadcolignan A,compounds 2,4,and 7—11 were isolated from this genus for the first time.At a concentration of 50μmol/L,compounds 4 can significantly reduce the release of NO in LPSstimulated RAW264.7 cells,with inhibition rates of 64.45%.
