摘 要:目的:建立浙麦冬组织培养快繁体系,为其规模化生产提供技术支撑。方法:以‘浙麦冬1号’嫩叶基部为外植体,研究植物生长调节剂对诱导愈伤组织、不定芽分化和不定根形成的影响,并进行了炼苗与移栽。结果:‘浙麦冬1号’愈伤组织诱导最适培养基为MS+KT 2.0 mg/L+NAA 0.5 mg/L+蔗糖30.0 g/L+琼脂2.5 g/L+活性炭2.0 g/L,诱导率为92.22%;愈伤组织分化不定芽最适培养基为MS+KT 4.0 mg/L+NAA 0.5 mg/L+蔗糖30.0 g/L+琼脂2.5 g/L+活性炭2.0 g/L,不定芽分化率达88.89%;最适生根培养基为1/2 MS+NAA 0.5 mg/L+蔗糖30.0 g/L+琼脂2.5 g/L,生根率为96.67%;组培苗经驯化,移栽2周后成活率为89.00%。结论:建立了‘浙麦冬1号’组织培养快繁体系。Objective:To establish tissue culture and rapid propagation system of Ophiopogon japonicus from Zhejiang,so as to pro-vide certain theoretical basis and technical support for its scientific and large-scale production.Methods:Taking the leaf base of Zhejiang Ophiopogon japonicus No.1″as an explant,the effects of plant growth regulators on inducing callus,clump bud differentiation and adventitious root formation were studied,and the seedlings were refined and transplanted.Results:The optimal medium for callus in-duction of Zhejiang Ophiopogon japonicus No.1″was MS with 2.0 mg/L KT,0.5 mg/L NAA,30.0 g/L sugar,2.5 g/L agar,2.0 g/L activated carbon,the induction rate was 92.22%;The optimal medium for callus differentiation adventitious buds was MS with 4.0 mg/L KT,0.5 mg/L NAA,30.0 g/L sugar,2.5 g/L agar,2.0 g/L activated carbon,and the adventitious bud differentiation rate was 88.89%.The optimal rooting medium was 1/2 MS with 0.5 mg/L NAA,30.0 g/L sugar,2.5 g/L agar,and the rooting rate was 96.67%;The survival rate of tissue culture seedlings was 89.00%after two weeks of domestication.Conclusion:The rapid propagation technology of Zhejiang Ophiopogon japonicus No.1″tissue culture is established.
