北国壁钱和华南壁钱的鉴别研究

摘　　要：目的:筛选两种药用蜘蛛北国壁钱Uroctea lesserti Schenkel,1936和华南壁钱Uroctea compactilis L.Koch,1878有鉴别意义的特征,完善传统动物药的鉴定方法。方法:采用形态分类学方法观察和比较两个物种的形态特征,同时考察线粒体DNA细胞色素C氧化酶亚基I(mtDNA-COI)基因序列的特征和遗传距离。结果:华南壁钱和北国壁钱原动物形态、药材及其网巢彩色特征图及比较表显示二者存在明显差异。两种壁钱的COI序列长度为630~681 bp,在NJ树中可各自聚成一支,并与外群完全分开。所得到的COI基因片段序列上传至GenBank数据库,分别获得北国壁钱的登录号OP358932和华南壁钱的登录号OP363143。结论:该研究为两种壁钱药材的真伪鉴定提供了重要的实验依据。Objective:To screen the distinguishing characteristics of two medicinal spiders,Uroctea lesserti and Uroctea compactilis,and improve the identification methods of traditional animal drugs.Methods:Morphological characteristics of the two species were ob-served and compared by morphological taxonomic methods,and mitochondrial DNA cytochrome C oxidase subunit I(mtDNA-COI)gene sequence characteristics and genetic distance were investigated.Results:The original animal morphology,color maps and tables of the medicinal materials and their netting and nest showed obvious differences between Uroctea compactilis and Uroctea lesserti.The length of COI sequence of the two species was 630 to 681 bp,which could be clustered into one in NJ tree and completely separated from the out-group.The obtained COI gene sequence was uploaded to GenBank database,and the entry number OP358932 of Uroctea lesserti and OP363143 of Uroctea compactilis were obtained,respectively.Conclusion:This study provides an important experimental basis for the authenticity identification of two kinds of Chinese medicine.
