摘 要:目的:采用UPLC和数字影像技术比较生地黄和熟地黄的差异。方法:采用相似度评价、配对样本t检验、Wilcoxon符号平均秩检验、正交偏最小二乘判别分析(OPLS-DA)、聚类分析等多元统计分析方法对生地黄、熟地黄的特征图谱和色度值进行分析。结果:建立的生地黄、熟地黄特征图谱分别确定了18、20个共有峰,其中色谱峰19(5-羟甲基糠醛)、20为炮制新增成分,且生地黄与熟地黄特征图谱中色谱峰3~9、11~18的峰面积差异具有统计学意义。OPLS-DA结果表明色谱峰1、2、4、19是导致生地黄和熟地黄差异的主要因素。色彩图像分析中,熟地黄与生地黄的色差ΔE^(*)为16.47~31.11,可明显识别生品与炮制品,色度值L^(*)、a^(*)、b^(*)、E^(*)均可作为生地黄与熟地黄的鉴别依据之一。结论:该研究建立的生地黄、熟地黄特征图谱方法稳定、可靠,结合色度值可用于区分生地黄及其炮制品熟地黄。Objective:To compare the differences between Rehmanniae Radix and Rehmanniae Radix Praeparata by using UPLC and digital imaging technology.Methods:The chromatograms and chromaticity value of Rehmanniae Radix and Rehmanniae Radix Pra-eparata were analyzed by using multivariate statistical analysis methods,such as similarity evaluation,paired sample t-test,Wilcoxon sign average rank test,orthogonal partial least squares discriminant analysis(OPLS-DA),cluster analysis,etc.Results:18 and 20 com-mon peaks were identified by the established UPLC characteristic spectrum of Rehmanniae Radix and Rehmanniae Radix Praeparata,respectively.Among them,peaks 19(5-hydroxymethylfurfural),20 appeared after the processing of Rehmanniae Radix.The peak areas of peaks 3 to 9 and peaks 11 to 18 were significantly different between Rehmanniae Radix and Rehmanniae Radix Praeparata.The OPLS-DA results showed that the peaks 1,2,4,19 were the main factors in the difference of Rehmanniae Radix and Rehmanniae Radix Praeparata.TheΔE^(*)between Rehmanniae Radix and Rehmanniae Radix Praeparata was 16.47 to 31.11,indicating that raw and pro-cessed products could be clearly identified.The L^(*),a^(*),b^(*)and E^(*)could be used as one of the distinguishing bases between Reh-manniae Radix and Rehmanniae Radix Praeparata.Conclusion:The UPLC characteristic spectrum method of Rehmanniae Radix and Re-hmanniae Radix Praeparata established in this study is stable and reliable,and it can be used to distinguish Rehmanniae Radix and Re-hmanniae Radix Praeparata by combining the chromaticity value.
