摘 要:目的:观察放疗后大鼠腮腺损伤的过程中B细胞淋巴瘤-2(Bcl-2)和Bcl-2相关x蛋白(Bax)在腮腺细胞的表达,探讨阳中求阴方在放疗引起的腮腺损伤中的防护机制.方法:将60只Wistar大鼠随机分为空白组、放疗组及联合组,各20只.空白组不进行放疗,放疗组及联合组在全麻状态下进行放疗照射,联合组于放疗前1周开始给予阳中求阴方,空白组与放疗组给予等剂量生理盐水.分析空白组、放疗组和联合组放疗后腮腺重量、细胞调亡率及Bcl-2、Bax mRNA的表达量.结果:放疗组和联合组腮腺重量低于空白组,细胞凋亡率高于空白组,差异有统计学意义(P<0.01);联合组腮腺重量高于放疗组,细胞凋亡率低于放疗组,差异有统计学意义(P<0.05).联合组和放疗组腮腺细胞中的Bcl-2 mRNA表达量低于空白组,Bax mRNA表达量高于空白组,差异有统计学意义(P<0.01);联合组Bcl-2 mRNA表达量高于放疗组,Bax mRNA表达量低于放疗组,差异有统计学意义(P<0.05).结论:阳中求阴方对放疗引起的腮腺损伤有保护作用,可抑制腮腺细胞凋亡,其机制可能通过抑制Bax mRNA的表达,同时上调Bcl-2 mRNA的表达来实现.Objective:To observe the expression of B-cell lymphoma-2(Bcl-2)and Bcl-2 associated x protein(Bax)in parotid cells of rats after radiotherapy,and to explore the protective mechanism of Yangzhong Qiuyin formula in parotid gland injury induced by radiotherapy.Methods:Sixty Wistar rats were randomly divided into blank group,radiotherapy group and combined group,with twenty rats each.The blank group was not given radiotherapy,and the radiotherapy group and the combined group were given radiotherapy under general anesthesia.The combined group was given Yangzhong Qiuyin formula one week before radiotherapy,and the blank group and the radiotherapy group were given the same amount of normal saline.The parotid weight,apoptosis rate and Bcl-2 and Bax mRNA expression levels were analyzed in blank group,radiotherapy group and combined group.Results:The weight of parotid in radiotherapy group and combined group was lower than that in blank group,and apoptosis rate was higher than that in blank group,the difference was statistically significant(P<0.01);the weight of parotid gland in combined group was higher than that in radiotherapy group,and the apoptosis rate was lower than that in radiotherapy group,the difference was statistically significant(P<0.05);the expression of Bcl-2 mRNA in parotid cells in combined group and radiotherapy group was lower than that in blank group,and the expression of Bax mRNA was higher than that in blank group,the difference was statistically significant(P<0.01);the expression level of Bcl-2 mRNA in combined group was higher than that in radiotherapy group,and the expression level of Bax mRNA was lower than that in radiotherapy group,with statistical significance(P<0.05).Conclusion:Yangzhong Qiuyin formula can protect parotid gland damage caused by radiotherapy and inhibit parotid cell apoptosis,which may be achieved by inhibiting the expression of Bax mRNA and upregulating the expression of Bcl-2 mRNA.
